Objective: Using LC-MS analysis research on plasma biomarkers 0f children with exacerbated asthma,studying on the pathogenesis of bronchial asthma from the perspective of metabolic pathway;By analyzing the changes of plasma biomarkers in children with cold and heat asthma on exacerbated asthma and to exploring the metabolic biomarkers of exacerbated bronchial asthma in children from the perspective of metabolic pathways.Methods:The experiment was divided into bronchial asthma group and normal control group,by collecting 48 cases of with cold asthma and 14 cases of hot asthma,30 cases of heathy children as normal control group;plasma samples were detected by liquid chromatography-mass spectrometry(LC-MS),Principal Component analysis and Partial least square-discriminant analysis were used to screen the plasma differential metabolites of bronchial asthma children and control group,bronchial asthma with cold and heat asthma group,at last,using the MetaboAnalyst platform analyze the possible metabolic pathways.Results:Clear distinction between the groups,different metabolites and metabolic pathways can be identified(1)Acute exacerbation of bronchial asthma in children and healthy controls plasma samples can be distinguished,cold asthma group compared with control group,33 and 17 different metabolites can be find under positive and negative ion mode,heat asthma group compared with control group,34 and 16 metabolites different metabolites can be screened.(2)Compared bronchial asthma group with control group,further speculates that metabolic pathways are related to the pathogenesis of bronchial asthma,including sphingolipid metabolism,glycerophospholipid metabolism,arachidonic acid metabolism,retinol metabolism,arginine and proline metabolism,phosphoinositide metabolism,glycine,serine and threonine metabolism,valine,Leucine and isoleucine metabolism,taurine metabolism.(3)Compared with the cold asthma group and the heat asthma group,18 metabolites can be found,and conclude that there may be two related metabolic pathways,which are glycerolphospholipid metabolism pathway and peanut acid metabolism pathway.Conclusion:Plasma metabolites in bronchial asthma exacerbation of children and health children with significant differences,which may be related to the pathogenesis of bronchial asthma.By analyzing the different plasma samples of biomarkers and metabolic pathways can reveal the syndrome relationship of exacerbation bronchial asthma between cold group and heat group.
