Effect Of Dahuangfuzi Decoction On Severe Acute Pancreatitis Induced Intestinal Motility Disorder In Rat

Objective: To explore the mechanism of Dahuangfuzi decoction on intestinal motility dysfunction by observing the effects of Dahuangfuzi decoction on serum motilin,Cajal interstitial cells and motilin receptor in rats with intestinal motility disorders induced by severe acute pancreatitis.Methods: SPF male SD rats were randomly divided into control group(n = 6),SAP group(n = 6)and Dahuangfuzi decoction group(n = 6).According to the previous animal model preparation method,rat model of severe acute pancreatitis was prepared based on the rat body weight(1ml/ kg)by injecting sodium taurocholate(4%)into retrograde cholangiopancreatic duct.The rats in the SAP group were given 2ml normal saline(37 ℃)enema at 12 and 24 hour after model making.The treatment group was given 2ml Dahuangfuzi decoction(37 ℃)enema at 12 and 24 hour respectively.The control group only open belly,gently turn the pancreas several times,then close the abdomen.After modeling for 48 hour,the rats in each group were anesthetized,and then the abdominal aorta blood samples were taken.Serum inflammatory cytokines IL-1β and motilin were detected by ELISA.The pathological changes of pancreas and ileum were observed by the HE staining.Schmidt method was used to evaluate the pancreatic tissue in each group.The ileum tissue was evaluated by Chiu’s method.The expression of c-kit and motilin receptor protein in interstitial cells of cajal(ICC)in ileum tissue was detected by immunohistochemistry.Results: Dahuangfuzi decoction could significantly decrease the level of serum endotoxin(P <0.05),amylase(P <0.05)and IL-1β(P <0.05);At the same time,the level of motilin can be increased(P<0.05)by Dahuangfuzi decoction;Dahuangfuzi decoction can increase the number of ICC-expressing protein c-kit and motilin receptor MTL-R immunoreactive cells in SAP group(P <0.05).Conclusion: The structure of intestine in SAP was damaged.The concentration of plasma motilin and the number of ICC in ileum was decreased.The number of the motilin receptor and ICC in smooth muscle of ileum was decreased.The intestinal motility was disrupted.Dahuangfuzi decoction can inhibit the inflammatory response,regulate the secretion of MTL,repair damaged intestine and restore the number and function of ICC,which can improve the intestinal motility disorders in rats.
