Obesity is mainly caused by abnormal accumulation of adipose tissue in the body.It is one of the most serious health problems in the world.Adipose tissue can be divided into white adipose tissue?WAT?that stores energy and energy-consuming brown adipose tissue?BAT?.Since the role of VEGFB gene in lipid metabolism was discovered, people began to pay more attention to the role of VEGFB gene in adipose tissue differentiation and energy metabolism. We know that VEGFB gene can form two subtypes of VEGFB186 and VEGFB167due to the different cleavage patterns. At present, there are few studies on the two subtypes alone, but this article is mainly to explore the role of this subtype of VEGFB186 in lipid metabolism. We linked the AP2 promoter to VEGFB186 and transplanted it into mice to specifically express it in adipose tissue to study the effect of VEGFB186 on lipid metabolism.First we observed its effect on adipose tissue development. We found that:VEGFB186 is in the wild Even if the type is specifically overexpressed in the adipose tissue of VEGFB-deficient mice, the body weight will be reduced;the weight of white adipose tissue will be reduced and the brown adipose tissue will be increased by weighing the adipose tissue; The size of white adipocytes became smaller and more abundant in HE staining, and the number of lipid droplets in brown adipocytes became smaller. The amount of adipose tissue development-related factors in each genotype mouse was measured. It was found that when VEGFB18686 gene was over When expressed,it can increase the expression of brown adipose tissue-related factor PGC-1?and decrease the expression of Leptin,a white adipose tissue-related factor, thereby regulating adipose tissue development, reducing white adipose tissue, and increasing brown adipose tissue.Reduced the accumulation of adipose tissue and increased energy consumption,to make mice become thinner. In energy metabolism,it was found that when VEGFB186 is overexpressed in wild mice,it can increase the expression of FATP3 and FATP4 in adipose tissue. When it is overexpressed in VEGFB-deleted mice,it can increase the expression of FATP4 in adipose tissue.Increased energy metabolism;subsequent measurement of body temperature also found that when VEGFB186 was overexpressed, its body temperature was relatively high;then its glucose tolerance was measured and it was found that when VEGFB186 is overexpressed,it can rapidly perform glucose metabolism, inhibit the increase of glucose, and increase its Glucose tolerance. Therefore,we can find that when VEGFB186 is over-expressed,it can regulate the development of adipose tissue and energy metabolism,so that the accumulation of adipose tissue is reduced, and the energy metabolism is accelerated,so that the body weight of mice is reduced. And can improve glucose tolerance. It is proved that VEGFB186 can play a role similar to VEGFB in lipid metabolism. In the future treatment of obesity-related diseases, this effect of VEGFB18686 has important reference value for the development of targeted drugs in the future.
