Metabolic syndrome(MS)is a group of syndromes with central obesity,hyperlipidemia,hypertension,diabetes,or fasting blood glucose.Chinese medicine using the overall concept and the combination of individual treatment methods in the prevention and treatment of metabolic syndrome has achieved remarkable results.Modern medicine on the pathogenesis of metabolic syndrome,the understanding of the pathogenesis remains to be further studied,and test the clinical efficacy of traditional Chinese medicine treatment of metabolic syndrome also need effective animal experimental technical support,and therefore meet the clinical characteristics of metabolic syndrome syndrome combined with animal models Become an effective experimental means to study this problem.The existing MS animal models are mostly induced by diet-induced superimposed drugs or directly selected gene-deficient animal modeling.The model often reflects only the characteristics of a single disease,and there is a certain gap with the actual human factors and omnivorous eating habits The Previous studies on the MS model are often more concerned about the model of the model,and its pathophysiological characteristics of the evolution of no relevant research.Disease and animal model as a new theoretical innovation,its standardization and standardization there are many theoretical and technical issues need to be resolved step by step.Based on this,this study is based on the animal model of metabolic syndrome syndrome,and tries to establish a comprehensive evaluation system of metabolic syndrome and animal model similar to the clinical features and explore the evolution rule of MS syndrome.1A preliminary study on the metabolic syndrome model of C57 mice induced by high-fat and high-sugar and high-salt dietC57 mice were randomly divided into normal group and model group,10 rats in each group and normal group.The model group was given high fat and high sugar and high salt diet for 4 weeks.Mice were weighed weekly and blood pressure,blood glucose,grip,and pulse were measured after 4 weeks.The results showed that there was no significant difference in body weight,blood glucose,blood pressure,grasping force and pulse data between the normal group and the model group.According to the experimental results,this formula feed failed to succeed after 4 weeks of experiment.Prompt to replace the formula feed,adjust the experimental program.2 Screening of high fat and high sugar and high salt formula feed for inducing metabolic syndrome rat modelAccording to the diet characteristics and modeling requirements of rodents,two new formula feed a and b were selected,and Wistar rats were given for 4 weeks respectively.After 4 weeks,the rats in the normal group fed with basal diet Comparison of the Exploratory Study of Rat Model of Metabolic Syndrome Combined with High Fat and High Glucose and High Salt Diet in Rat ModelFour weeks old Wistar rats were randomly divided into normal group and model group(24 rats in each group).The rats were given normal diet and high fat and high sugar diet.The rats were sacrificed at the 4th,8th and 12th week respectively.The body weight,blood glucose,blood pressure,blood lipid,liver histopathology Visceral fat coefficient,lee's index and TCM syndrome index.After 4 weeks,the body weight of the model group was higher than that of the normal group(P<0.05),and the mild fatty liver-like changes were seen in the pathological sections of the liver.After 8 weeks,the body weight and lee's index,the perirenal fat coefficient,serum total cholesterol(P<0.01).The fat density of low density lipoprotein(LDL)and testis was higher than that of normal group(p<0.05),and the fatty liver disease was increased.After 12 weeks,The levels of serum total cholesterol(TC),low density lipoprotein(LDL),kidney and testicular fat were significantly increased(P<0.01),lee's index and body weight(p<0.05),high density lipoprotein)Decreased(P<0.05),liver pathological sections showed moderate fatty liver-like lesions.At 4 weeks,the model group decreased the open field activity,the heart rate increased,the swimming endurance,the respiratory rate,the pulse frequency,the pulse amplitude decreased and the change was continued until the end of 12 weeks(P<0.05).The results of dynamic observation showed that the pathogenesis of MS showed an increasing trend with the increase of time.The obesity signs of abdominal obesity were the first,and the indexes of blood lipid and the change of fat in the liver were changed in the middle stage of the experiment.(P<0.05).At 12 weeks,the rats showed a phlegm-dampness syndrome model(lighter)).4 Evaluation of the Effect of Metabolic Syndrome on the Spleen Qi Model of Metabolic SyndromeRespectively,using three different methods to extract metabolic syndrome(Astragalus 30g Ligustrum lucidum 12g Coptis 5g Trichosanthes 9g Alisma 12g Apocynum 12g wild chrysanthemum 15g),administered for 4 weeks,the model group and the normal group comparison,Pharmacodynamics evaluation.After 4 weeks,there were significant differences in body weight,testicular fat coefficient,open field movement time,pulse amplitude and swimming endurance in the administration group and model group.Metabolic syndrome has an improved effect on the model of spleen qi deficiency in rats,and it also demonstrates the scientificity of metabolic syndrome model of rat spleen deficiency induced by high fat and high sugar and high salt diet.
