Objective:To explore the protective effects and possible mechanism of Xiangxi selenium-riched kiwi fruit root polysaccharide on CCl4-induced acute liver injury mice.Method:Use ultrasonic-assisted way to extract kiwifruit root polysaccharide;Mice were randomly divided into six groups including blank group,model group,polyene phosphatidyl choline group?195.4mg/?kg·d??,low-dose kiwifruit root polysaccharide group?40 mg/?kg·d??,middle-dose kiwifruit root polysaccharide group?80mg/?kg·d??and high-dose kiwifruit root polysaccharide group?160 mg/?kg·d???1h after the last gavage on the seventh day,The blank group was received 10ml/kg olive oil by intraperitoneal injection,While other groups were administered with CCl4 solution by intraperitoneal injection to establish acute hepatic lesion model.After 16 hours,the mice were anesthetized and took blood by excise the eyeballs.Contents of serum alanine aminotransferase?AST?and aspartate aminotransferase?AST?were measured immediately.Then took the liver and detected the contents of SOD and MDA in the liver.Immunohistochemical method was used to measure the expression of TNF-?and IL-1?in the liver.Results:?1?The data were analyzed by one-way ANOVA.Compared with the blank group,the levels of ALT and AST in the model group were significantly increased?P<0.01?,suggested that the model was established successfully.Compared with the model group,the levels of ALT and AST in the three doses of XiangXi selenium-riched kiwifruit root polysaccharide groups and polyene phosphatidyl choline group significantly decreased?P<0.05?.?2?Compared with the blank group,SOD in the liver homogenate of the model group significantly decreased?P<0.01?,while MDA significantly increased?P<0.01?.Compared with the model group,SOD in the liver homogenates of the high-dose group,middle-dose group,and polyene phosphatidyl choline group was significantly increased?P<0.05?.The MDA content decreased significantly?P<0.05?.?3?According to the observation under the optical microscope,the liver tissue of the blank group was almost normal,while the liver tissue of the model group was obviously observed:cytoplasm looseness,balloon-like change,punctate necrosis,inflammatory cell infiltration,some of them even see the bridge necrosis,large necrosis.Compared with the model group,the above pathological changes of the liver cells of the drug intervention groups all improved to a certain extent,among which the high-dose polysaccharide group improved most obviously.?4?Immunohistochemistry analysis:Compared with the blank group,the protein expression of TNF-?and IL-1?in model group was significantly increased?P<0.01?.Compared with the model group,the levels of TNF-?and IL-1?in the liver tissue of high,medium and low dose groups of polysaccharide group were lower than those in model group?P<0.05?.However,statistical analysis found that there was no significant statistical difference between the high,medium and low-dose polysaccharide groups and polyene phosphatidyl choline group.?P>0.05?Conclusion:1.Xiangxi selenium-riched kiwifruit root polysaccharides can protect experimental acute liver injury caused by CCl4.2.Xiangxi selenium-rich kiwifruit root polysaccharides can downregulate TNF-?and IL-1?protein expression.in acute liver injury mice which caused by CCl4.3The protective effect of Kiwifruit root polysaccharides may be associated with antioxidant free radical damage,regulation of cytokines,and relieves mitochondrial oxidative stress.
