Objective:Firstly,High-throughput sequencing was used to sequence circulating miRNAs in patients with heart failure,and then qRT-PCR was used to verify the expression of circulating miRNA-423-5p in patients with heart failure,using the receiver operating curve(ROC)assess the value of circulating blood miRNA-423-5p as a molecular marker for the diagnosis of heart failure.Methods:(1)A total of 144 heart failure patients(including 78 patients with ischemic heart failure and 66 patients with non-ischemic heart failure)were enrolled from July 2014 to November 2015 in Shenzhen Sun Yat-sen Cardiovascular Hospital.Normal controls 53 cases.In the high-throughput sequencing stage,8 cases of ischemic heart failure,8 cases of non-ischemic heart failure and 8 cases of healthy subjects were selected;70 cases of ischemic heart disease and non-ischemic heart failure were selected during the qRT-PCR validation stage.Cardiomyopathy 58 patients with heart failure and 45 healthy people.(2)Specimen collection and preservation: Anticoagulated venous blood was prepared using 0.11 M sodium citrate anticoagulant.After 2000 g of centrifugation,the plasma was retained after 15 minutes of centrifugation,and the temperature was kept at-80 degrees Celsius for later use.(3)High-throughput sequencing was used to screen the samples for sequencing.The results of the two heart failure groups were compared with the normal control group to calculate differential expression.P<0.05 was a statistically significant difference.(4)RT-PCR was used to detect circulating blood miRNA-423-5p and internal reference U6 expression levels.(5)Statistical analysis: SPSS 19.0 statistical software was used for statistical analysis of data,and ROC curve and area under the curve(AUC)were used to evaluate the diagnostic value of miRNA-423-5p in heart failure.Results:(1)Compared with the normal group,miRNA-423-5p increased 4.176 times in ischemic heart failure,and miRNA-423-5p increased 14.264 times in non-ischemic heart failure.(2)The AUC value of miRNA-423-5p is 0.776,which is of moderate diagnostic value.Conclusion:Circulating miRNA-423-5p in heart failure are heightened,miRNA-423-5pmay be a potential molecular biomarker for diagnosing heart failure.
