The PTPN18 Expresses In Endometrial Carcinoma And Its Correlation With Clinicopathological Features Study

Objective: The purpose of this study is to study the expression of PTPN18 in endometrial cancer,reveal its correlation with clinicopathological features,and lay an experimental foundation for subsequent biological function tests of endometrial cancer cells.Methods:1.60 cases of paraffin sections and clinical information of cancerous and non-cancer tissues from patients with gynecologic endometrial cancer in Hainan General Hosital between April 2016 and April 2017 were collected.2.The expression of PTPN18 in tissue samples was detected by immunohistochemistry and its correlation with clinicopathological features was analyzed.3.PSC61492-1 lentivirus interfering vector for silencing PTPN18 gene was constructed and tested for virus titer.The recombinant plasmid was then transfected into HEC-1-A cells.The relative expression of PTPN18 m RNA in cells was detected by q RT-PCR.Results:1.Immunohistochemistry results showed that the expression rates of PTPN18 in cancerous and non-cancer tissues of patients with endometrial cancer were 68.33%(41/60)and 48.33%(29/60),respectively.The difference was statistically significant.2.Analysis of the correlation between PTPN18 and clinicopathological features revealed that the positive rate of PTPN18 expression in endometrial cancer tissues with stage II-III,depth of invasion ? 1/2,lymph node metastasis,and poorly differentiated endometrial cancer tissue was significantly higher than that of stage I,infiltration depth<1/2 muscle layer,no lymph node metastasis and highly differentiated endometrial cancer tissue.The positive rate of PTPN18 expression in each group was compared and statistically analyzed,with statistical significance,P<0.05.3.Successfully constructed the PSC61492-1 lentivirus interfering vector,detected the number of virus drops as 1.0X109TU/m,and successfully transfected HEC-1-A cells.The results of q RT-PCR showed that relative to the control group,the PTPN18 gene of the experimental group was relatively The expression level was 0.208,and the knockdown efficiency reached 79.2%.Conclusions: 1.The expression of PTPN18 in endometrial carcinoma is significantly increased.It may serve as an detection marker for endometrial carcinoma.2.The expression of PTPN18 is related to FIGO stage,depth of muscle infiltration,lymph node metastasis and histological grade,and may be a predictor of malignancy of endometrial carcinoma.
