Background:Cervical cancer ranks second among female lethal tumors,which seriously threatens the health and lives of women.The continuous infection of human papilloma virus(HPV)is associated with the onset of cervical cancer.About 99.7%of cervical squamous cell carcinomas and more than 94%of cervical adenocarcinomas can be detected with high-risk HPV infection.With the popularity of cervical cancer screening,more and more patients with cervical precancerous lesions and cervical cancer can be timely diagnosis and treatment.However,after standardized treatment of cervical cancer patients,there are still about 22.6%to 35%of patients with recurrence or metastasis,which is the main reason leading to death of cervical cancer patients.Therefore,early detection and diagnosis and treatment of cervical precancerous lesions and cervical cancer,it is of great significance to effectively evaluate the prognosis of cervical cancer patients.Many researchers hold the opinion that HR-HPV viral load is another key independent factor leading to the development of cervical cancer in addition to persistent infection with HR-HPV.But there is still much controversy about the correlation between the HR-HPV viral load and the degree of cervical lesions.hPygo2(human Pygopus 2,hPygo2)is a nuclear protein.As one of the major effector proteins in the Wnt signaling pathway,hPygo2 has been found to be involved in the development of many human tumor diseases.hPygo2 has biological functions such as regulating tissue development,transcriptional activation of downstream genes,and regulation of chromatin remodeling processes.Tzenov et al.found that hPygo2 protein was over-expressed in CINIII and cervical squamous cell carcinoma tissue,and through a series of experiments,they proved that E7 displaces Rb from Elf-1 and results in Elf-1-mediated hPygo2 expression.There is a close relationship between hPygo2 and HPV E7.However,there is no relevant research on the correlation between hPygo2 and HR-HPV viral load and the relationship between clinicopathological features and prognosis of cervical cancer.Therefore,this study examined the expression of hPygo2 in LSIL,HSIL and cervical cancer tissues,analyzed the relationship between hPygo2 and HR-HPV viral load,and preliminary explored the role of hPygo2 expression and HR-HPV viral load in the development of cervical disease.The correlation between hPygo2 expression and clinical pathological features of cervical cancer was analyzed.The relationship between hPygo2 and prognosis of cervical cancer patients was explored,which provided new ideas for the prognosis evaluation of cervical cancer.Objective:By examining the expression of hPygo2 in LSIL,HSIL and cervical cancer tissues,the relationship between hPygo2 and HR-HPV viral load was analyzed,and the role of hPygo2 expression and HR-HPV viral load in the development of cervical diseases was preliminary explored.To investigate the relationship between hPygo2 expression and clinicopathological features and prognosis of cervical cancer patients.Methods:1.From January 2010 to June 2014,109 cases of cervical cancer,89 cases of HSIL,49 cases of LSIL and 44 cases of normal cervix were selected from the gynecological departments of Zhujiang Hospital of Southern Medical University.2.Hybrid Capture 2 testing was used to detect the patient's HPV viral load.3.Expression of hPygo2 in different cervical tissues:Immunohistochemistry was used to stain normal cervix,LSIL,HSIL and cervical cancer tissues.According to the cell staining intensity and the number of positive cells,the result was judged.4.SPSS 21.0 statistical software was used to process all the experimental data.The differences in the expression of hPygo2 in different cervical tissues and the differences of HR-HPV in different cervical lesions groups were performed using non-parametric tests of multiple independent samples.Positive rate The comparison was performed using the x2 test and Spearman rank correlation analysis was used to analyze the correlation coefficient.Kaplan-Meier method was used for survival analysis and OS,DFS curve mapping,and Log-Rank test was used for further comparison.Differences in survival rates between the two groups;univariate and multivariate analyses were analyzed using Cox regression models.P<0.05 was considered statistically significant by a two-sided test.Results:1.The appearance of brown-yellow granules as hPygo2 positive cells was observed under a microscope.From LSIL and HSIL to cervical cancer tissues,the number of hPygo2 positive cells gradually increased and the staining intensity increased.The expression of hPygo2 in each group was statistically significant.2.The positive rates of high-risk HPV in LSIL,HSIL,and cervical cancer tissues were 71.4%,96.6%,and 98.2%,the difference was statistically significant(x2=54.571,P<0.001).There was a positive correlation between HPV viral load and the degree of cervical lesions(Correllation Coefficient=0.320,P<0.001).3.In LSIL,HSIL and cervical cancer tissues,the expression intensity of hPygo2 increased with the increase in the high-risk HPV virus,and was positively correlated by Spearman rank correlation analysis(Correllation Coefficient=0.627,P<0.001).4.The positive expression of hPygo2 was related to the histological type,clinical stage,histological differentiation and lymph node metastasis in patients with cervical cancer(P<0.05).5.The overall survival time was longer in the hPygo2 negative group than in the positive group.Multivariate Cox analysis showed that hPygo2 positive expression,clinical stage,and lymph node metastasis were independent prognostic factors.Conclusion:1.hPygo2 is mainly expressed in the nucleus,and its expression in HSIL and cervical cancer tissues is significantly higher than that in normal cervical tissues and LSIL,suggesting that hPygo2 may be involved in the development of cervical precancerous lesions and cervical cancer.2.There was a positive correlation between the quantification of HR-HPV virus and the severity of cervical lesions.3.There was a positive correlation between hPygo2 expression and HR-HPV virus quantification.4.The expression of hPygo2 is closely related to the clinicopathological features such as clinical stage,histological type,histological differentiation and lymph node metastasis of cervical cancer.5.The expression of hPygo2 is closely related to the poor prognosis of cervical cancer.It is an independent prognostic factor for the overall survival of patients with cervical cancer.It has certain value in predicting the prognosis of cervical cancer.
