Objective: Objective to explore the mechanism of wound healing and the effect on wound healing by using medical radiation protection spray combined with red light treatment.Methods: A total of 32 female SD rats of clean grade,buttock skin of each rat was produced by beta ray linear accelerator(45Gy)single irradiation,the size of 40 mm * 30 MM,caused by acute deep II degree burn wounds of beta ray skin at the same time,32 rats were randomly divided into four groups: combined treatment group,n=8.In rats with acute hip deep II beta ray after skin injury,topical medical radiation protective spray 0.1ml/,2 cm,2 times / day,and combined with red light treatment,light source distance wound 20 cm,2 /d,each 30min/ irradiation time,red light wavelength(640 + 10)nm.Single spray group,n=8,the wound appeared alone after topical medical radiation protective spray 0.1ml/,2 cm,2 times / day.The red light group and n=8 were used alone.After the emergence of the wounds,they were treated by red light alone.The light source was 20 cm from the wound surface,2 times /d,each irradiation field 30min/ times,and the red light wavelength was(640 + 10)nm.In the control group,n=8,after the appearance of the wound,the wound was treated with saline.In the wound of rats appeared after 0,1,2,3,4,5 weeks,the gross observation and changes of rat wound spirit;to observe the pathological changes of wound tissue and VEGF expression in rats by light microscope and electron microscope;the dynamic changes of SOD and MDA concentration in molecular biology detection method to detect the SOD concentration and MDA the rats in the blood.Results:(1)the wound healing time in the combined treatment group was 32.2 + 3.4 days,and the wound healing time in the spray group was 40.7 + 3.7 days.The healing time in the red light group was 39.1 + 4.1 days,and the healing time in the control group was 55.3 + 4.5 days.The combined therapy group and single spray group,red light alone group wound healing time was shorter than that in the control group,and the differences were statistically significant(P<0.05);combined treatment group wound healing time was faster than the two single treatment group,and the differences were statistically significant(P<0.05);single red group than single spray group the healing time is slightly faster,but there was no statistically significant difference between them(P>0.05).(2)Light microscope and electron microscope showed that the number of fibroblasts,epithelial cells and vascular endothelial cells in the three treatment groups was significantly higher than that in the control group.(3)Immunohistochemistry showed that: the combined treatment of 1 and 2,integral absorbance of VEGF group after 3 weeks(IA)was significantly higher than that of the other three groups,the differences were statistically significant(P<0.05);integral absorbance two single treatment group VEGF(IA)value is higher than the control group,and the difference is statistically significant(P<0.05)however,integral absorbance of VEGF between two single treatment group(IA)value was not statistically significant(P>0.05).(4)The concentration of SOD in the United treatment group after 1,2,3,4 and 5 weeks of blood and skin tissues,significantly higher than the other three groups,and 2,3,4,5 weeks MDA was significantly lower than the other three groups,and the differences were statistically significant(P<0.05);the concentration of SOD two single drug treatment group after 1,2,3,4 and 5 weeks of blood and skin tissues,significantly higher than the control group,and 2,3,4,5 weeks,the concentration of MDA was significantly lower than the control group,the differences were statistically significant(P<0.05);while the SOD concentration between two single medication treatment group after 1,2,3,4 5 weeks,blood and skin tissues and in 2,3,4,5 weeks MDA concentration had no significant difference(P>0.05).Conclusion: The combination of Medical ray protective spray and red light therapy can promote the healing of the wound of skin injury of beta ray skin in rats and shorten the time of wound healing.And all were better than the single use of Medical ray protective spray or red light therapy.
