Research On Expression Of LncRNA Zeb1-AS1 In Osteoarthritis Chrodrocyte And Normal Chrodrocyte

Object: osteoarthritis is a chronic disease of joint,as a character of disabling and reducing quality of life.The major clinic features are joint pain,malformed appearance and movement disorder.And the main pathological features are cartilage degradation and osteoproliferation.Zeb1 is Zinc finger E-box-binding homeobox 1,research has dictated that Zeb1 is cartilage modifying gene.Zeb1-AS1 is Zeb1 antisense transcript 1,a kind of lncRNA,which is a primary transcript over 200 nt RNA has complex biological functions and homology of mammals,it plays an important role in biological tissue and organ development.The purpose of this study was to find the expression of lncRNA zeb1-as1 in osteoarthritis chondrocytes and normal chondrocytes.Methods: primary cell culture technique was used to develop knee osteoarthritis and normal knee cartilage cells.The expression levels of Zeb1-as1 and Zeb1 in osteoarthritis chondrocytes and normal chondrocytes were measured by Real-time Quantitative PCR.WB.Result: The expression levels of Zeb1-as1 and Zeb1 in knee osteoarthritis were higher than those in normal chondrocytes.Conclusion: Zeb1-as1 and Zeb1 were up-regulated in the cartilage cells of osteoarthritis of the knee.
