Objective: The aim of this study was to explore ZEB1-AS1 expression levels and evaluate its function and mechanism in bladder cancer cells.Methods: The expression of LncRNA ZEB1-AS1 was examined by quantitative real-time PCR(qRT-PCR)in 36 pairs of bladder cancer tissues(and corresponding non-tumor tissues)and bladder cancer cells,and the bladder cancer cell lines were screened.Using Specific si-ZEB1-AS1 to construct interference of cell line and using pcDNA3.1 ZEB1-AS1 to construct stable overexpression cell line.Then,the function of ZEB1-AS1 expression in the progression of bladder cancer cells were determined by molecular experiments.CCK-8 and colony formation assay were employed to detect the function of ZEB1-AS1 on proliferation.Flow cytometric analysis of apoptosis was applied to test the function of ZEB1-AS1 on apoptosis,western blot was used to detect the expression of apoptosis-related protein caspase 3.Wound healing assay,transwell assay were applied to exam the function of ZEB1-AS1 on cell invasion and migration.Western blot assay was used to determine the expression level of EMT-related proteins.The expression of ZEB1 in 36 cases of bladder cancer(corresponding to normal adjacent tissues)was detected by qRT-PCR.The expression of ZEB1 in bladder cancer cells after overexpression and interference with ZEB1-AS1 was detected by qRT-PCR and Western blot,then analyzing the correlation between ZEB1-AS1 and ZEB1.Results: LncRNA ZEB1-AS1 was found up-regulated in bladder cancer tissues(compared to adjacent non-tumortissues)and was almost overexpression in bladder cancer cells(compared with normal urothelial cell line).Increased lncRNA ZEB1-AS1 expression can significantly promoted tumor cells proliferation,inhibited cells apoptosis,facilitated cells invasion and migration and contribute to the formation of EMT phenotype.While down-regulated ZEB1-AS1 could obviously inhibit cells proliferation,promote cells apoptosis,suppress cells metastasis and inhibit EMT.There was a positive correlation between the expression levels of ZBE1-AS1 and ZEB1 in bladder cancer tissues.And ZEB1-AS1 overexpression can promote the expression of ZEB1 and interfere with ZEB1-AS1 can inhibit the expression of ZEB1,which indicating that ZEB1 may be a potential target for ZEB1-AS1 in bladder cancer.Conclusions: These results indicated that lncRNA ZEB1-AS1 was associated with tumor progression and tumorigenesis in bladder cancer cells,could be used as a new target for the prognosis of bladder cancer...
