Objective: 1)The predisposition to ischemic stroke(IS)might involve in interactions of several genes and environmental factors.Polymorphism has been speculated to be and extensively investigated as a risk factor for IS.The present study was aimed to evaluate the single nucleotide polymorphisms(SNPs)of homocysteine methylation reaction genes as risk factors for ischemic stroke in Uyghur population.Methods: 367 IS patients and 1202 controls were genotyped for investigated genes by Polymerase Chain Reaction-Restriction Fragment Length Polymorphism(PCR-RFLP)method.Results:.Even missense variants Ala222 Val and Glu429 Ala of the 5-Methylenetetrahydrofolate reductase(MTHFR)gene,Asp919 Gly of the MTR gene,Lys350 Arg,Ile22Met,Arg415 Cys of the MTRR gene and Arg239 Gln of BHMT gene were selected to examine their relationships with IS.The frequency of CC and CA genotype and C allele of the Glu429 Ala polymorphism of the MTHFR gene were higher in IS group(7.5%,44.8% and 29.9%)than that of control group(6.1%,36.4% and24.3%).Under a dominant model of minor-allele C,CC and CA genotype carriers showed an increased risk in comparison with AA with an OR of 1.485(95% CI 1.170-1.883,P<0.05)in the IS group.The difference was significant after adjusting for age,gender,smoking,drinking,obesity,hypertension status,total cholesterol and triglyceride levels.No significant findings of the rest of six polymorphisms with IS was observed.Conclusion: Our results suggest that Glu429 Ala polymorphism of MTHFR gene is involved in the prevalence of IS in Uyghur population and C allele of Glu429 Ala polymorphism might be the risk factor of IS in Uyghur.
