Objective:The number of adult patients seeking orthodontic treatment has increased in recent years.About half of the woman in the world will face alveolar bone absorption caused by postmenopausal osteoporosis,which brings out some problems such as root absorption loose teeth,relapse in or after orthodontic tooth movement.A large number of clinical and basic studies have confirmed that Osteoking can effectively promote bone foRMation.In this project,the orthodontic tooth movement models of osteoporosis SD rats were established,and then the SD rats were gavaged by Osteoking.We Detected jaw density of rats in different time nodeS,mineral salt content,and measured the distances of teeth movement in rats.Ultimately through the MicroCT detection,slicing of HE staining and immunohistochemical experiment of bone morphogenetic protein 2(BMP-2),We evaluated the remodeling effect of Osteoking on the alveolar bone around the moving tooth.This study intends to explore a new path that the yunnan ethnic medicine Osteoking promotes alveolar bone remodeling in osteoporosis,so that we can provide some eXperimental basis for the clinical prevention of the root absorption and recurrence of adult movement teeth.Methods:Part ?,establishment of osteoporosis animal model.In the experiment,45 3-month-old SD rats were selected and randomly divided into 3 groups.The control group(blank group),the model group(removal of the ovary)and the drug group(removal of the ovary and drug intervention).All rats underwent initial bone density measurements,The model group and the drug group were excised with bilateral ovaries and sutured.After 5 days of the removal of the ovary,the rats in the drug group were gavaged with Osteoking.After 10 years,All rats underwent bone density measurements again,The bone mineral density,jaw mineral density and mineral salt content were compared in three groups.Part II:The establishment of teeth movement model and measurement of tooth movement distance in rats.45 SD rats were divided into three groups.Control group(teeth movement),model control group(removal of ovary and teeth movement)and drug group(removal of ovary,teeth movement and drug intervention).After the three groups established the teeth movement model,The distances of teeth movement were measured at 7d,14d and 21d of orthodontic force.And the rats were killed at the above time.Part ?:The study on the remodeling of alveolar bone around the moving teeth of rats.the mandibular tissue In the second part of the experiment was removed,the movement teeth and its surrounding alveolar bone of the rat were removed.Part of them were used for microCT detection,and the other part is for tissue sections.Through MicroCT examination,HE staining,immunohistochemistry,the alteration of alveolar bone and root absorption were observed in rats among different groups.Results:1.The first part of experimental research showed that,the bone mineral density and jaw mineral density were measured in the three groups,which were higher in the 10th week than in the first week;In the control group and in the operation group,there were differences in bone mineral density and jaw mineral density between the first week and the 10th week(P<0.05);In the feeding group and the operation group,there were differences in bone mineral density and jaw mineral density between the first week and the 10th week(P<0.05).The bone salt content in the three groups also increased,but the difference was not statistically significant(P>0.05).2.The second part of experimental research showed that,in the same time,the teeth movement distances were measured,The displacement distance in the model control group was longer than which in the control group(P<0.05),and The displacement distance in the drug group was shorter than which in the model control group(P<0.05).3.The third part of experimental research showed that,in the detection of MicroCT,The number,thickness and separation degree of bone trabecula were analyzed.Compared with the control group,bone microstructure degeneration in the control group.Compared with the model control group,The microstructure of the bone in the drug group showed better.In the sections of HE staining,we observed that osteoblasts of the alveolar bone were more in the drug group.There was a certain level of root absorption in the model control group.The immunohistochemical sections manifested that the positive expression rate of BMP-2 in the drug group was higher than which in the control group.Conclusions:1.The rat model of osteoporosis was established by resection of bilateral ovaries.Osteoking can delay the loss of bone mass in female ovariectomized rats,and is effective in the treatment of osteoporosis.2.The teeth movement model of rats were successfully established by using the teeth moving device of spring and ligation wire.The speed of tooth movement could be accelerated due to osteoporosis.The intervention of Osteoking could slow down the rapid teeth movement in osteoporosis.3.Osteoking could promote the remodeling of the alveolar bone around the moving teeth in osteoporosis rats,osteoblasts were active,the expression of BMP-2 increased.In addition,the teeth movement of osteoporosis rats could accelerate the root absorption.The intervention of Osteoking could reduce root absorption to a certain extent.
