Objective: Geniposide,a main iridoid glucoside component of gardenia fruit,has been known to exhibit antioxidant,anti-inflammatory,inhibition of platelet aggregation,prevention of atherosclerosis and other important therapeutic activities.Relative researches have proved that activating the Nrf2/HO-1 affect the results such as antioxidative,antiatherogenic and others.High level lipoprotein(a)in plasma is the independent risk factors of As.A growing number of researches have shown that the level of lipoprotein(a)in plasma is related to the quantity and the molecular weight of apolipoprotein(a).Thus,the main strategy of anti-atheroscleros is at present is how to surpress the apo(a)level in plasma.In this experiment,we study the mechanism how Geniposide have effect on intercellular apo(a)expression in hep G2 cells.Methods: Hep G2 cells were cultured in DMEM supplemented with 10%FBS at 37 °C in a humidified incubator under 5% CO2? In order to have the synchronization growth,the hep G2 cells were cultured in fresh serum-free medium for 6 hours before studying the confluence.Cells were treated with different concentrations geniposide(50,100,200 ?g/ml)for 24 h,and the cells were maintained at 37?C in 5%CO 2 for 0,6,12,and 24 h while using200?g/m L geniposide.Using Western blotting and Real Time-PCR methods detect the apo(a)expression in protein and m RNA levels.Results: In the solution culture,increasing the concentration of the geniposide from 50ug/ml to 200ug/l,compared with control group,the reduction of apo(a)m RNA and protein levels can accompany in hep G2 cells.We observed that the 100-200ug/l concentration have the biggest effectives.Incubating the cells from 0-24 h,we observed that the apo(a)m RNA and protein levels,compared with the control group,have declined,and the obvious changed occurred in 48 hours.Using the inhibitor or small interfering RNA(si RNA)inhibit HO-1expression,the effect that Gp decrease apo(a)levels have disappeared.Conclusions: 1: In Hep G2 cells,Gp inhibit the expression of apo(a)with dose and time dependent2: Gp downregulate the expression of apo(a)via Nrf2/HO-1 signaling.
