Objective: We are determined to investigate the effect of total flavonoids of Mosla henryi on anti-hepatic fibrosis and to explain its mechanism of anti-liver fibrosis.Methods: Fifty-six Sprague Dawley rats were selected and 10 of them were selected as the control group,other rats were injected intraperitoneally with 40% CCl4 olive oil to make liver fibrosis model.Pathological findings prompted the successful establishment of liver fibrosis rat model after 8 weekend,the 31 surviving rats were then randomly divided into model group(n = 16)and treatment group(n = 15).The treatment group were given MSF 160mg/kg of Mosla henryi,the model group was given the same dose of saline during the same period.Except for the blank control group,the remaining two groups were given 40% CCl4 olive oil by intraperitoneal injection of 1mg / kg until the end of the experiment.After 12 weeks,rats were killed(10 control groups,12 model groups,14 treatment groups),serum aminotransferase(ALT,AST)was detected by automatic biochemical analyzer.Four indicators of hepatic fibrosis(HA,LN,PC?,?-C)were detected by radioimmunoassay,and we take liver tissue to HE and Masson staining,liver histopathological changes were observed under light microscop,and semi-quantitative liver fibrosis score,the expressions of PDGF-BB and CTGF in the liver of each group were detected by IHC and WB.Results:1?The serum levels of ALT,AST,HA,LN,PCIII,and IV-C in the model group were significantly higher than those in the control group(P<0.01),the treatment group was significantly lower than the model group(P <0.01).2?The serum levels of semi-quantitative liver fibrosis score?the expression levels of PDGF-BB and CTGF in liver tissue in the model group were significantly higher than those in the control group(P<0.01),the treatment group was significantly lower than the model group(P <0.01).Conclusions:1?MSFcan reduce the levels of ALT,AST,HA,LN,PC?,?-C in serum of rats with hepatic fibrosis and then reduce the degree of hepatic fibrosis in hepatic pathological tissues of liver fibrosis rats.2?The anti-hepatic fibrosis of MSF may be related to the down-regulation of PDGF-BB and CTGF expression in hepatic tissues of rats with hepatic fibrosis.
