Objective: Research effects of Astragaloside IV on the expression of CTGF and FN in cardiac fibroblasts.To explore the molecular mechanism of Astragaloside IV in inhibiting myocardial fibrosis,as to provide theoretical basis for Astragaloside IV used for clinical purposes.Methods: Cardiac fibroblasts(CFs)were isolated from the hearts of1-to 3-day-old Neonatal Wistar rats.Second passage cardiac fibroblasts were used for all experiments.MTT method was used to determine the optimal concentration of drug intervention.According to the different experimental groups,the corresponding intervention was conducted: the control group: no intervention was added;Empty carrier group: the carrier of empty plasmid;sh-CTGF group;Astragaloside IV group;TGF-β1 group,sh-CTGF+TGF-β1 group,sh-CTGF+Astragaloside IV group.And CFs were identifed by their specifc marker vimentin.RT-PCR and Western blot were used to identify the expression of the genes CTGF and FN in cardiac fibroblasts.Results:1.The extracted cardiac fibroblasts were spindle shaped,diamond-shaped,polygonal and other shapes,and the positive rate of cell was >95%.2.There is no significant difference of the mRNA levels of CTGF and FN between the control group and the empty carrier group.Theseresults suggest that empty vectors have no effect on gene expression.3.There is significant difference of the mRNA levels of CTGF and FN between the control group and sh-CTGF group,which suggested that the transfection is successful and the intervention is effective.4.There is significant difference of the mRNA levels of CTGF and FN between the control group and TGF-β1group.These results suggest that TGF-β1could stimulate the expression of CTGF and FN mRNA in myocardial fibroblasts.5.There is significant difference of the mRNA levels of CTGF and FN between the control group and TGF-β1group,which suggest that TGF-β1 could stimulate the expression of CTGF and FN mRNA in myocardial fibroblasts with sh-CTGF intervention.6.There is significant difference of the mRNA levels of CTGF and FN between the control group and ASIV group,which suggested that ASIV could inhibit the expression of CTGF and FN mRNA in cardiac fibroblasts.7.There is significant difference of the mRNA levels of CTGF and FN between the sh-CTGF group and sh-CTGF+TGF-β1group,which suggested that ASIV could inhibit the expression of CTGF and FN mRNA in cardiac fibroblasts with sh-CTGF intervention.Conclusion:1.Astragaloside IV can reduce the expression of CTGF and FN incardiac fibroblasts.2.TGF-β1 can stimulate the expression of CTGF and FN in cardiac fibroblasts.3.The expression of CTGF and FN in cardiac fibroblasts stimulated by TGF-beta 1 was positively correlated.4.Astragaloside IV may inhibit the expression of CTGF and FN by inhibiting TGF-β-smads.5.Astragaloside IV may inhibit myocardial fibrosis and improve myocardial remodeling,and play a protective role in the heart by reducing the expression of FN and CTGF in cardiac fibroblasts.
