Purpose:To observe the effects of Jie Du Tong Luo Fang on the expression of CTGF and TGF-beta 1 in myocardial fibrosis rats,and to explore the mechanism of Jie Du Tong Luo Fang mediated TGF-beta 1-CTGF pathway in the intervention of myocardial fibrosis.Method:36 male SD rats of 8 weeks old were randomly divided into control group(group CK),model group(group MF),low,medium and high dose of Jie Du Tong Luo Fang group and ACEI group(n = 6).5mg/kg weight control group received saline,other groups were given 5mg/kg body weight of isoprenaline hydrochloride(ISO)by intraperitoneal injection,myocardial ischemia model,from the beginning on the day,Jie Du Tong Luo Fang is low,medium and high dose group every day,rats were given Jie Du Tong Luo Fang concentration of 1g/ml TC suspension in 10,20 and 30ml/kg body weight ACEI group received intragastric administration,the concentration of 0.5mg/ml Kato Pury suspension 10ml/kg ig.Group CK was given distilled water.After modeling 3W,24 h after the last administration were at the apical sites of rats by MASSON staining to calculate the collagen volume fraction(CVF),immunohistochemistry was used to detect the expression of TGF-1 and CTGF protein in myocardial tissue,to detect the expression of TGF-beta 1 and CTGFmRNA RT-PCR.Result: the expression of CK in myocardial tissue contrast group1.MF group TGF-beta 1 and CTGF protein and mRNA expression increased significantly(P<0.05)comparing MF group2 Jie Du Tong Luo Fang high medium and low dose group myocardial TGF-beta 1 and CTGF protein and mRNA were significantly lower(P<0.05)Jie Du Tong Luo Fang3 high medium and low dose group the comparison between the two groups had no significant difference(P>0.05).Conclusion: Jie Du Tong Luo Fang may slow down the development of myocardial fibrosis by inhibiting the expression of TGF-1 and CTGF protein.
