Objective:In this study,we detected the expression levels of four microRNAs,including mi R-16,miR-21,miR-12,and miR-221 in normal healthy subjects and patients with stroke at different time points(respectively 1 day,7 days,14 days,and 6 months after stroke happening).The purpose of this study is to find micro RNA expression levels in plasma related to the mRS of stroke patients and to explore indicators that may predict the mRS of patients with stroke.Methods:68 acute stroke patients at different time points(respectively 1 day,7 days,14 days,and 6 days after stroke happening)were recruited.Besides,we collected peripheral venous blood samples of 31 healthy control.Plasma were divided within 4h.The total RNA was extracted from plasma and then reversed transcripted into cDNA.The expression levels of mi R-16,miR-126,mi R-221,miR-21 and U6 as internal reference were detected in plasma of healthy controls and acute stroke patients by real-time quantitative PCR.Relative expression levels of microRNAs in the plasma were calculated by2~-(??Ct??Ct) method.The mRS score was used to evaluate the prognosis of stroke patients at 180 days.The stroke patients with mRS score?2 were considered as the good prognosis group and stroke patients with mRS score>2 were considered to be the poor prognosis group.We statistically analyzed the correlations between the expression levels of these target microRNAs with prognosis of stroke patients.Results:1.The relative expression levels of plasma mi R-16,mi R-21,miR-126,and miR-221 in the stroke patients group at 1 day from stroke onset were significantly higher than healthy controls(p<0.01).2.The relative expression levels of plasma mi R-16,mi R-21,miR-126,and mi R-221 at 1 day from stroke onset were not significantly different among different prognosis groups(p>0.05).3.The relative expression levels of plasma mi R-16,mi R-21,miR-126,and miR-221 were significantly different among different prognosis groups at7 day from stroke onset.Relatively expression levels in poor prognosis were higher(p<0.001).4.Relatively expression levels miR-16 and miR-126 in poor prognosis groups at 14 day from stroke onset were higher(p<0.05),while the relative expression levels of miR-21 and miR-221 were not significantly different among different prognosis groups(p>0.05).5.By ROC curve analysis,we found that the relative expression levels in the plasma of miR-16,miR-21,and mi R-126 at 7 day from stroke onset could be a prognostic marker of stroke with medium diagnostic efficiency.6.By ROC curve analysis,we found that the plasma relative expression levels of miR-16,and miR-126 at 14 day from stroke onset could be a prognostic marker of stroke with medium diagnostic efficiency.7.ROC curve analysis showed that every combination of miR-16,miR-21,and miR-126 in plasma at 7 days from stroke onset showed a high predictive value for the prognosis of stroke.8.The relative expression levels of plasma mi R-16,mi R-21,miR-126,and miR-221 were unchanged in stroke patients at different time.Conclusions:1.The relative expression of plasma miR-16,miR-21 and miR-126 at 7day from stroke onset could be used to predict the mRS.The higher the expressions of miR-16,miR-21,and miR-126 are,the higher the mRS and the prognosis of stroke patient was.The predictive value of the combined indexes of each two of plasma miR-16,miR-21,and miR-126 at 7 day from stroke onset were higher.2.There was no correlation between the expression level of mi R-221 in the plasma and mRS.
