Background:Stroke is one of the leading causes of death and disability in humans,and ischemic stroke accounts for about 80%of strokes.At present,although the 1-year survival rate of cerebral ischemic patients exceeds 80%,up to 70%of patients still have neurological function defect due to missed acute-phase treatment timing or poor results even after acute-phase treatment.Therefore,it is necessary to explore a new method to improve the neurological function during the recovery stage from cerebral ischemia.In recent years,bone marrow mesenchymal stem cells(BMSCs)have become a research hotspot in this field due to their role in tissue repair and regeneration.Many experimental studies have shown that BMSCs transplantation can significantly improve the neurological function of rats with acute ischemic brain damage and improve their prognosis.However,does the transplantation of BMSCs improve the neurological function of cerebral ischemia recovery?There are still few relevant reports.Axonal growth factor Nogo-A can inhibit axonal regeneration and axonal plasticity,and plays an important role in the nervous system,and it may closely relate to acute cerebral ischemic injury.Does it play a role in cerebral ischemia in recovery?However,there are few studies on this topic.Therefore,this study aimed to investigate the effect of BMSCs transplantation in rats with cerebral ischemia in recovery stage and its effect on Nogo-A signaling pathway,so as to further explore the mechanism of BMSCs transplantation in improving the neurological function of cerebral ischemia rats during recovery stage.Aim:To investigate the effect of bone marrow mesenchymal stem cells transplantation on recovery of neurological functions and expression of Nogo-A in recovery stage cerebral ishchemia rats.Methods:Bone marrow mesenchymal stem cells were isolated and cultured by the whole bone marrow adherence method.Thirty-six SD rats were randomized into sham operation group,ishchemia group and BMSCs transplantation group with 12 rats in each group..The ischemic model was established with line embolism to block the middle cerebral artery in the ishchemia group and BMSCs transplantation group,After 21 days built the recovery stage cerebral ischemia model,1ml of BMSCs(3×106)transplanted into the rats in the BMSCs transplantation group,and the sham operation group and ishchemia group were given the injection of the same amount of PBS.After the treatment of 14 and 28 days,Rats were evaluated the neurological functions by mNSS scores.The brain infarct sizes were tested by TTC staining,the pathological alteration tested by HE staining,and The expression of Nogo-A were determined by immunofluorescence.Results;After the treatment of 14 and 28 days,as compared with those in the control group and ischemia group,the mNSS score in BMSCs group were significantly reduced(14 d:7.50±0.55 vs.6.17±0.75;28 d:7.33±0.52 vs.5.67±0.82).What's more,Rats in the BMSCs group had significantly smaller brain infarct sizes and scar tissues than the control group and ischemia group(14 d:26.32%vs.31.38%;28 d:21.68%vs.21.71%).The expression of Nogo-A in the BMSCs group were significantly lower than those in the control group and ischemia group(14 d:33.67%vs.39.33;28 d:25.67%vs.30.33%).The HE staining showed that the cell damage of BMSCs group was milder than ischemic group,and the scar tissue was reduced.Conclusion:The transplantation of BMSC can improve the neurological function of the recovery stage cerebral ishchemia rats,and it may work by down-regulating the expression of Nogo-A.
