Effects Of Bone Marrow Mesenchymal Stem Cells’ Transplantation On The Recovery Of Acute Spinal Cord Injury Rat Neurological Functions And The Expression Of Nogo-A

Abstract/Summary:
Objective: To investigate the effects of intravenous administration of bone marrow-derived mesenchymal stem cells(BMSCs)into rat tail vein on the recovery of neurological functions and the formulation of Nogo-A after acute spinal cord injury.Method: Bone marrow-derived mesenchymal stem cells were harvested and purified following adherence technique. The rat model of acute spinal cord injury was established with modified Allen’s method.In short ninety-six female adult Sprague-Dawley rats were randomized into four groups,and each group is twenty-four.There are Sham-operated group,SCI group,Vehicle group and BMSCs-treated group. About 5×106 cells were injected into tail vein at 10 minutes after SCI.The vehicle group was injected with the same dose of PBS. BMSCs were labeled by CFSE before the transplantation. At 1d、3d,7d and 14 d after SCI,the rats neural functions were estimated by using the Basso-Beattie-Bresnahan(BBB) before sacrificed. At 1d、3d,7d and 14 d after SCI,the Nogo-A was estimated by immunohistochemical method and by reverse transcription polymerase chain reaction(RT-PCR) after sacrificed.Result: In the BMSCs-treated group,the CFSE marked positive cells were catched surround the spinal cord injury region. The rats neural functions in BMSCs-treated group were better than the SCI group and Vehicle group, at 7d、14d after SCI,and nogo-A around the injury area was evaluated significantly cut at 3d、7d、14d after SCI.(all P<0.01). There was no obvious distinction of neural functions in rats’ spinal cord 1d and 3d post SCI among the SCI group 、 Vehicle group and BMSCs-treated group.Conclusion: After acute spinal cord injury, the rat neural functions can be obviously improved after the BMSCs’ transplantation and its mechanism and theory was interfixed to descend nogo-A expression.
