BackgroundsChinese medicine polysaccharide has obvious effect in antitumor and has the advantages of safety,side effects and other advantages.Recent studies have shown that the anti-tumor effect of compound Chinese medicine polysaccharide is obviously better than that of traditional Chinese medicine monosaccharide.The combined use of several traditional Chinese medicine polysaccharide can effectively control the growth of tumor.In this study,the oral liquid formulation of Poria Cocos polysaccharide is composed of polysaccharide of Lentinusedodes,Ganodermalucidum and Poria cocos etc.In this study,the content of polysaccharide in the oral liquid of Poria Cocos polysaccharide was determined and the mechanism of antitumor was studied.Objectives1.The determination of polysaccharide content in the oral liquid of Poria Cocos polysaccharide.2.The evaluation of the antitumor mechanism of the oral liquid of Poria Cocos polysaccharide.MethodsABSTRACT1.The determination of polysaccharide in the oral liquid of Poria Cocos polysaccharide by phenol-sulfuric acid method.2.The tumor model was established by mouse S180 sarcoma model.Western blot method was used to analyze the protein contents of B-cell lymphoma-2(Bcl-2),cysteinyl aspartate specific proteinase-3(caspase-3)and cysteinyl aspartate specific proteinase-9(caspase-9)in thymus,spleen and liver tissue of mice in each experimental group.Hematoxylin eosin(HE)staining method was used to analyze the morphology of thymus,spleen and liver cells in each experimental group.Immunohistochemical method was used to analyze the protein contents of Bcl-2,caspase-3 and caspase-9 in thymus,spleen and liver tissue of mice in each experimental groupResults1.The determination of polysaccharide content in the oral liquid of Poria Cocos polysaccharideThe polysaccharide content of the oral liquid of Poria Cocos polysaccharide was 17 mg/mL.2.The experiment of the antitumor pharmacological mechanism research on the oral liquid of Poria Cocos polysaccharide.2.1 Western blot experimentCompared with the blank group,the expression of Bcl-2 protein in the thymus,spleen and liver in other groups increased significantly(p<0.01).Compared with the model group,the expression of Bcl-2 protein in the control group and the high dose group decreased significantly(p<0.01),and the expression of Bcl-2 protein in the high dose group was lower than it in the control group,but there was no statistical difference.The expression of caspase-3 and caspase-9 in the thymus,spleen and liver was the same.Compared with the blank group,the expression of protein in the control group and the high dose group increased significantly(p<0.01).Compared with the model group,the protein expression in the control group and the high dose group increased significantly(p<0.01).Compared with the model group,the high dose of the protein expression was higher than it,but there was no statistical difference.2.2 Hematoxylin eosin(HE)staining experimentThe cells in the blank group were arranged closely in the thymus,spleen and liver,there were slightly little edema and normal space.The cells in the model group were irregular crinkled and the intercellular space became larger,and there were large cell necrosis and fatty degeneration in the tissue.The control group had better cell status,there were no necrosis and fatty degeneration.The cells in the high dose group were no obvious pathological changes,the cells condition was good,and the state was more normal than the control group.2.3 Immunohistochemistry experimentThe maximum of the integral optical density value(IOD)of Bcl-2 protein in the thymus,spleen and liver was the model group,and the minimum was the blank group.Compared with the blank group,the model group increased significantly(p<0.05).Compared with the model,the high dose group decreased significantly(p<0.05),and the IOD value of the high dose group was lower than the control group,but there was no statistical difference.The maximum of the integral optical density value(IOD)of Bcl-2 protein in the thymus,spleen and liver was the high dose group,and the minimum was the model group.Compared with the blank group,the high dose group was significantly higher(p<0.05).Compared with the model group,the high dose group was significantly higher(p<0.05),and the IOD value of the high dose group was higher than the control group,but there was no statistical difference.Conclusions1.The phenol-sulfuric acid method can be used to determine the content of the polysaccharide in the oral liquid of Poria Cocos polysaccharide.2.The oral liquid of Poria Cocos polysaccharide can maintain the cell morphology of thymus,spleen and liver in S180 tumor mice.3.The mechanism underlying the antitumor effects of the oral liquid of Poria Cocos polysaccharide involved inhibition of Bcl-2 expression and activation of caspase-3 and caspase-9 expression.So the cell was apoptosis.
