Naringin and narirutin are flavonoids isolated from Cirtri Pericarpium,which is a product from the peels of oranges.They have lots of biological activities.In this work,the anti-allergic activities of flavonoids were done in vivo and vitro.In vitro,the releasing of?-HEX of RBL-2H3 cell line was applied to evaluate the anti-allergic effects.As the results show,comparing with naringin,the inhibitory effects of the narirutin on the releasing of?-HEX was stronger.The results showed that narirutin has better inhibitory activity than naringin.In vitro,the OVA induced allergic asthma model was applied to investigate the effect of narirutin in vivo.The results show that narirutin has anti-allergic effects by regulating of T cell response.The details are as follows:1.The inhibition effects of narirutin and naringin on releasing?-HEX of RBL-2H3 cells.(1)The effects of narirutin and naringin on the viability of RBL-2H3 cells were investigated by MTT.The results showed that narirutin and naringin had no effects on the viability activity of RBL-2H3 in the concentration range of 0.3-100?mol/L.(2)The effects of narirutin and naringin on the release of?-HEX were investigated.Narirutin and naringin could significantly inhibit the releasing of?-HEX from activated degranulation of RBL-2H3 cells induced by IgE monoclonal antigens in the concentration range of 0.3-30?mol/L,and showed a concentration-dependent tension,the results indicating that the inhibitory activity of narirutin was better.(3)The effects of narirutin on releasing of IL-4 in the supernatant of RBL-2H3cells were investigated by ELISA assay,the results showed that it could significantly inhibit the release of IL-4 from activated degranulation RBL-2H3 cells in the concentration range of 3-30?mol/L.2.The anti-allergic effects of narirutin in vivo.Based on the OVA induced BALB/c mouse model of allergic asthma,the effects of narirutin was evaluated.(1)Narirutin could significantly decrease the level of the IgE and IgG1 level of allergic asthma mice induced by OVA.(2)Narirutin could significantly suppress the content of Th2 typical cytokines IL-4,and IL-5 increase Th1 typical cytokines IFN-?in bronchoalveolar lavage fluid by OVA induced allergic asthma mice;(3)Narirutin can affect the percentage of T cells in spleen lymphocytes about CD4~+and CD8~+T cells in spleen lymphocytes.(4)Narirutin can decrease the p44/42 and JNK pathway activated by OVA.In conclusion,narirutin could inhibit the releasing?-HEX in vitro.Narirutin can reduce the level of antibody in serum and decrease the level of typical inflammatory factor secreted by Th2 in bronchoalveolar lavage fluid and increase the cytokines of Th1.The ratio of T cells in spleen lymphocytes about CD4~+and CD8~+was affected.Narirutin can decrease the p44/42 and JNK pathway activated by OVA.It had anti-allergic activity in vivo.
