Objective:The aim of this study was to establish a food allergy animal model in2~3weeks BALB/C mouse with OVA,to explore different doses of OVA on food allergy model,and to observe the values of CPG-ODN on Th1/Th2 and ma st cell chemotactic protein1(m MCP-1)in serum of young mouse with food allerg y.To analyze significance of m MCP-1 as food allergies new detecting-mark in cli nical use.Method:Forty female BALB/C mice(2~3Week)were used to establish the food allergy animal model. They were randomly divided into five groups: Control gr oup, OVA induced food allergy groups(20 μg group;50 μg group;100 μg grou p) and CPG-ODN Intervention group(50μg OVA+CPG-ODN).The sensitization w as divided into basic and challenge phases.Basic sensitization by intraperitoneal O VA at day0,14,then stimulated by oral-feeded the same dose every other day at d ay17-29. Intervention group was pretreated with CPG-ODN intraperitoneally befo re OVA inhalation challenges every time.The allergic symptoms were observed an d scored. Blood sample was collected from each mouse. Th1, Th2 cells and seru m OVA-Ig E and m MCP-1 levels were determined. Jejunal tissue was taken for h istopathological examination.Result:1.Preparation of an animal model of food allergy in mice successfully:①According to the symptom score system: control group did not appear allergy symptoms; the score of20μg group was 22(2.75);50μg group was 31(3.88); 100μg group was 19(2.38);② Serum specific Ig E levels compared with the control group, OVA-Ig E content ofthree dose groups were significantly higher than the control group(≥mean + 3S), the difference was statistically significant(P = 0.001 <0.01);③The pathologic expression of the mice with food allergies compared with the control group, the presence of a large number of lamina propria lymphocytes and a few eosinophils invasion, Hemangiectasis obviously. The degree of inflammatory cell infiltration and varying degrees of hemangiectasis were different between three doses group, which was remarkable in 50μg group, which was alleviated in the CPG-ODN group.2.The serum levels of IL-4/ IFN-γ,m MCP-1 of young mice.①Serum IFN-γlevels of sensitized mice :The IFN-γlevels of 20μg group, 50μg group were significantly lower compared with control group and there was statistically significant(P <0.05); 100μg group compared with the control group(P > 0.05); there were no statistically significant difference between 20μg group and 50μg, 100μg group(P >0.05);50μg group was lower than 100μg group(P <0.05);②Serum IL-4 levels of sensitized mice : The serum IL-4 levels of 20μg, 50μg, 100μg group were significantly increased compared with control group(P <0.01); serum IL-4 levels of 20μg group and 100μg were lower than 50μg group(P <0.05), 20μg group was higher than 100μg group(P <0.05);③Serum m MCP-1 levels of sensitized mice :The m MCP-1 levels of 20μg, 50μg,100μg group were significantly higher than the control group(P <0.05); 20μg group,100μg group were lower than 50μg group(P <0.05), 20μg group compared 100μg group(P >0.05, the difference was not statistically significant).3.After intraperitoneal injection of CPG-ODN, CPG-ODN group with 50μg group, the OVA-Ig E, IL-4, and m MCP-1 levels were decreased(P<0.05), IFN-γlevels increased(P<0.05, the differences were statistically significant).Conclusion1.The food allergy animal model with ovalbumin can be built in 2~3 weeks BALB / C mice.2.The effectiveness sensitized with OVA,50μg allergens is the best in the three dose groups, 20 μ g group followed, 100 μ g group worst. There is a certain dose-effect relationship with increasing doses of allergen.But it is the worst in 100 μg group,over-doses allergens can induce immune tolerance.3.The m MCP-1 involved in development of food allergy, which is expected to become one of the indicators of clinical detection with food allergies.4.CPG-ODN by inducing Th1-type immune response and thus reverse the imbalance of Th1 / Th2, inhibits food allergies in the immune response, plays an important role in immune regulation.
