With the economic development and improvement of living standards,human beings are paying more attention to healthy diet.A reasonable healthy diet and lifestyle are vital to improving homeostasis of body and extending lifespan.We define limiting of nutrients intake as dietary restriction.Autophagy is the basic event in the body.Dietary diet can induce autophagy,which participates in the regulation of lifespan.Cathepsin L(CPL-1),located in lysosomes,belongs to the cysteine protease family and is expressed throughout the nematode.In order to study the relationship between cathepsin L and autophagy,during dietary restriction,we constructed the eat-2 & Plgg-1gfp:: lgg-1 and cpl-1 & Plgg-1gfp::lgg-1 double mutant.By crossing the hermaphrodites of eat-2/cpl-1 with the male of Plgg-1gfp :: lgg-1,By Selfing,the homozygous with two mutants was generated.By observing the green fluorescence and gene sequencing,the homozygous double mutant was picked out.Finally,we detected the expression of cathepsin L and pumping rate in the double mutants.With the mutants,we studied the role of CPL-1 in autophagy during dietary restriction in C.elegans.Using the LGG-1 :: GFP nematodes,we found that,during different dietary restriction,autophagy was induced.Meanwhile,Lysosome number and the expression of cathepsin L increased.In order to clarify the relationship between autophagy and cathepsin L,we used cpl-1 mutant.We found that cpl-1 mutants were able to inhibit the increases of lysosome number during dietary restriction,resulting in the accumulation of LGG-1 :: GFP.Through inhibiting of substrate degradation autophagy,cpl-1 mutant depressed the extendion of lifespan during dietary restriction.The above results indicate that CPL-1 is involved in the regulation of autophagy during dietary restriction.In summary,this study found that during dietary restriction,CPL-1 is involved in autophagy-related regulation,inhibition of autophagy substrate degradation,thus affecting the extension of their life expectancy.CPL-1 plays a crucial role during dietary restriction,contributing to the regulatory mechanism of autophagy.
