| Objectives1)To examine the changes of bone mineral density and body fat in ovariectomized monkey with application of dual energy X-ray absorptiometry2)To verify the response of medication in ovariectomized monkeys by evaluation of bone mineral densityMethod20 healthy adult female rhesus monkeys of 8-12-year-old equivalent to40-45-year-old human were selected.The experiment was divided into four phases.Monkeys were examined by Hologic Company Dual-Energy X-ray bone density tester to detect the whole body,lumbar 3-6(L3-6),femoral neck,ward,trochanter,bone BMD and body fat content and whole-body muscle content in each phase of experiment.In the first phase DXA scans of 20 healthy monkeys were taken before ovariectomy and treatment and the whole body,lumbar 3-6(L3-6),femoral neck,ward,trochanter,bone BMD and body fat content and whole-body muscle content were evaluated.In the second phase after 3 monthsof first phase,the healthy monkeys were ovariectomized followed by evaluation.In the third phase after 3 months of second phase ovariectomized monkeys were supplied with medication and their readings were documented.Following fourth phase after three months of third phase,DXA was taken to evaluate the contents after treatment.All the reading from each phase was compared.Excel software was used to record the data and was processed for analysis using the statistical package for social sciences 20(SPSS)for windows.The data were compared by the paired t test.The test level is at 0.05 where p≤0.05 is statically significant.ResultThe total bone mineral density(BMD)and lumbar bone density(BMD)of the ovariectomized rhesus monkeys did not decrease significantly in the 0 and 3months.The total hip bone density,femoral neck density,the intertrochanteric bone mineral density,ward area bone density,the body fat content and the body fat percentage decreased significantly,and the difference was statistically significant.Compared with third months and sixth months,the total body fat content of the ovariectomized rhesus monkeys continued to decrease.The total body density,the bone density of the lumbar vertebra,the bone mineral density of the femoral neck,the bone mineral density in the intertrochanteric region and the bone mineral density in the ward area decreased significantly,and the difference was statistically significant.At Sixth month and ninth month,the total body fat percentage,whole body density,lumbar bone density,total hip bone density,bone density of the femur neck,bone mineral density of the intertrochanteric area and ward area showed an increasing trend,and the difference was statistically significant.Linear regression analysis of bone mineral density of lumbar vertebrae and femoral neck was performed bystandard regression coefficient where B=0.619 and 0.372.The linear regression analysis was used to calculate the standard partial regression coefficient where B=0.169,0.621 and 0.219 for the bone mineral density of the femoral neck,intertrochanteric region and ward area.The linear regression analysis was used to calculate the standard partial regression coefficient B=0.353,0.527,0.128 of the femoral neck,intertrochanteric region,ward area bone density and the bone mineral density of the femoral neck region from zeroth to third months.The standard partial regression coefficient B=0.380,0.663 and-0.028 were calculated by linear regression analysis on the bone mineral density and bone mineral density of the femoral neck region of the femoral neck,intertrochanteric region,ward area and the femoral neck region from third to sixth months.The B=-0.028 did not have statistical significance,and the rest were statistically significant.ConclusionAs we observed from the data,decreased BMD and fat contents after ovariectomy tend to increase significantly with the medication.However,during earlier medication,BMD at hip and lumbar both increased but fat content rather stayed decreased significantly at earlier stage of medication and only raised after later stage of medication suggesting less sensitivity of fat to the medication and more sensitive to adding bone density.Therefore,the positive response of the medication may be effective in treatment of Osteoporosis in human after estimating the correct dose and is with minimal adverse side effects. |
