Experimental Study Of Triple Traditional Chinese Medicine For Infusion Induced Phlebitis

Objective:Animal model of infusion phlebitis,To study the efficacy and mechanism of triple Chinese medicine preparation in the treatment of infusion phlebitis,to search for experimental evidence for treating phlebitis with triple Chinese medicine.Methods:1.Study on the anti-inflammatory effect of triple traditional Chinese medicine preparation:Kunming mice were randomly divided into 4groups:model group,solvent group,Hirudoid positive control group and triple Chinese medicine preparation group;two common acute inflammatory models of mouse ear swelling and toes swelling:Using croton oil to induce ear swelling in mice and toes swelling induced by carrageenan and then applied by external ointment,observed this two types of swelling degree.2.Study on the effect of triple Chinese medicine by mannitol induced ear phlebitis in rabbits,20 rabbits were injected with 20%mannitol intravenously,with a dose of 2.5ml/kg,5min infusion time,1 time/24 hr,and a total of 3 days,and intravenous infusion of another 4 rabbits by normal saline as a blank control group.On the third day,rabbits with successful phlebitis were randomly divided into four groups:model group,solvent group,positive control group or Hirudoid group,triple Chinese medicine preparation group,and were given drugs respectively,Observation of blood vessels and surrounding tissue by stereoscopic microscope.The 4^thday after administration blood and tissue were taken for histopathology and determine the measurement of tissue and serum inflammatory factors.The content of interleukin-6?IL-6?,tumor necrosis factor?TNF-alpha?and vascular endothelial growth factor?VEGF?in serum and tissue were measured by enzyme linked immunosorbent assay?ELISA?.Hematoxylin eosin staining?HE?was used to observe the histopathological changes of tumor tissues.Results:1.Compared with the model group,triple Chinese medicine preparation group has a significant statistical difference in the mouse auricular swelling induced by croton oil and toes swelling induced by carrageenan?P<0.05?.At the other hand,solvent group and Hirudoid positive control group have no statistical significant difference in this two common acute inflammatory models of mouse auricular swelling and toes swelling?P>0.05?.2.There were varying degrees of erythema,congestion and cord changes in rabbit auricular veins of the model group.Pathological findings also showed varying degrees of inflammatory changes.The success rate of the model was80%.Compared with the model group,triple traditional Chinese medicine group,solvent group and Hirudoid positive control group have no statistical significant difference in naked eyes and pathological findings and even related inflammatory factors?P>0.05?.There was no significant difference in the content of interleukin-6?IL-6?,tumor necrosis factor?TNF-alpha?and vascular endothelial growth factor?VEGF??P>0.05?.Conclusion:1.Triple Chinese medicine preparation group can obviously reduce the mouse auricular swelling induced by croton oil and toes swelling induced by carrageenan,indicating the anti-inflammatory effect of triple Chinese medicine group.It shows that triple Chinese medicine preparation has certain anti-inflammatory effect.2.Triple Chinese medicine preparation has no significant inhibitory effect on mannitol induced auricular phlebitis in rabbits.
