Investigation Of HSP60/HSP75 As Serum Biomarker Of Penumbra In Animal Model Acute Ischemic Stroke

Objective To extract specific protein biomarker both penumbra and serum and verify if HSP60/HSP75 were the specific serum biomarkers from ischemic penumbra of acute ischemic stroke or not.Methods Randomized conducting 30 male Sprague-Dawley(SD)rats into 5 groups(6 for each group)as middle cerebral artery occlusion(MCAO)models:sham surgery 0 h,surgery group 1 h,2h,3h,4h.Cerebral infarction models evaluated with neurological deficit score,Hematoxylin-Eosin(HE)staining.We investigated serum and penumbra samples by Itraq previously,and identified heat shock protein 60(HSP60)/HSP75 as candidate protein biomarkers according to the bioinformatics.Moreover,we verified the biomarkers which with synchronous expression trend utilizing test technique of Western blot(WB).Results IP plain tissues were stained by HE,microscopy indicated the normal,IP and IC tissues;IP showing neurons were slight edema,less nucleus stained deeply or apoptosis;IC tissue showing large amount of nucleus stained deeply,apoptosis and disappeared.From the WB test,HSP60/HSP75 expressions difference were significant in both SE and IP between sham and surgery groups(P<0.05).HSP60/HSP75 from IP were up-regulation both compared with sham group and IC section(P<0.05).Even though IP HSP75 up-regulation significantly compared to sham group;but SE HSP75 in surgery group no significant differed from sham group.Spearman correlation analysis indicated the difference of HSP60 or HSP75 between IP and SE were not significant.Conclusions HSP60/HSP75 expressions were significant in AIS model,and could be regarded as the specific protein biomarkers in IP;however,each or both of them couldn't be verified as serum specific biomarkers of IP.
