Objective: prostate cancer is one of the most common malignant tumors in the whole world.In China,the incidence of cancer and the cause of death of prostate cancer in our country are not as good as that of the European and American countries,but with the development of society and economy,the incidence of cancer is rising year by year,which threatens the health of men.In our study,we shown that prostate cancer is caused by multiple factors and genes,but we know little about the mechanism involved.With the rapid development of high-throughput microarray hybridization and sequencing technology,a large number of gene data and biological information are rapidly expanding.The NCBI(National Center for Biotechnology Information)integrated gene expression omnibus(GEO)is the largest public database for storing high throughput molecular data in the world.This study will explore and analyze the existing gene data in GEO database to find out the key genes and new therapeutic targets of prostate cancer.Material method: according to the standard process,the chip data in GEO are collected,processed and integrated,and the chip data are normalized and corrected with the compiled R language program.The results are pheatmap,Volcano Plot,protein Protein-protein interaction network(PPI).Combining with KEGG signnal pathway to analyse.The rule for differentially expressed geneswere according to: 1,the expression difference was more than 2 times or less than 0.5 times;2,P < 0.05.Results: according to the selected GEO database,GSE55945 group had 13 prostate cancer tissue chips,and 8 normal prostate tissue chips.There were 328 differentially expressed genes in the test samples,among which 233 genes were up-regulated and 95 genes were down regulated.The range of candidate genes was further reduced by thermography and volcano map analysis,and STRING online tools were used to analyze the genes and their encoded proteins.The results showed that the proteins and genes encoding them were mainly concentrated on 22 genes,such as RND3,CAV2,CAV1,CALM1,FLNA and so on.Further literature mining of these key genes revealed that FLNA gene may be associated with prostate cancer.Conclusion: the process of the development and transformation of prostate cancer can not be separated from the abnormal activation of related genes,cell adhesion and abnormal expression of genes related to migration.We use the GEO database to carry out the chip data retrieval and systematically analyze the differential gene discovery between normal prostate tissue and prostate cancer tissue.FLNA,DLX2 and other genes are closely related to the occurrence of prostate cancer,and provide a new potential gene site for the in-depth study of the pathogenesis of prostate cancer.GEO database analysis,as a large sample,small cost,simple analysis of the chip screening,through a variety of different data statistics and analysis measures,can be used for a huge number of people to carry out large-scale research on cancer genomics and to provide reference and basis for the follow-up research based on genomics.
