Objective:To study the mechanisn of Angelica blood tonic soup in inhibiting tumor growth of S180 tumor bearing mice.Methods:1.The model bearing tumor mice were constructed by implanting subcutaneously S180 tumor cells,then were divided into six groups:normal control group,model control group,positive control group and Angelica blood tonic soup high,medium and low dose groups?We would kill and record the weight including tumor,liver and spleen of mice and calculate tumor inhibition rate and index of immune organ after drugs treatment for ten days.2.HE staining was used to observe the morphology,size and arrangement of tumor tissues.3.In situ end labeling method(TUNEL method)was used to observe the apoptosis of tumor cells in different groups and calculate apoptosis index.4.Enzyme-linked immunosorbent assay(ELISA)was used to detect the serum il-2,TNF-alpha,and IFN-gamma in different groups.5.WESTERN-BLOT method was used to detect the effects of Angelica blood tonic soup on the jak-stat signaling pathway in tumor cells.Results:1.The Angelica blood tonic soup had obvious tumor inhibition effect on the S180 tumor-bearing mice,which were 36.53%in the high-dose group,38.00%in the medium dose group,and 27.42%in the low-dose group.2.Compared with the model group,the liver index and spleen index of the CTX group were significantly lower(P<0.05,P<0.01).The hepatic index and the spleen index of Angelica blood tonic soups were significantly higher than that in the CTX group(P<0.05,P<0.01).3.Tumor tissue morphology experimental results show that the tumor cells arranged sparse in Angelica blood tonic soup groups,and the proportion of tumor cells in the nucleus was decreased.The tissues of Angelica blood tonic soup groups were ruptured and empty,and some areas were replaced by non-tumor tissues.The apoptosis index(AI):the high dose group 18.25%,the medium dose group was 14.17%and the low-dose group was 9.44%.4.The serum il-2 content in S180 tumor-bearing mice was significantly increased in the Angelica blood soup group.5.The experimental results showed that the expression of p-stat3 could be significantly reduced in each Angelica blood soup group,and the effect of the high-dose group was the strongest.conclusion:Angelica blood tonic soup has obvious tumor suppressive effect on S180 tumor-bearing mice.It is possible to regulate the expression of TNF-alpha IL-2 and IFN-gamma in the serum of S180 tumor-bearing mice.Reduce the expression of p-stat3.
