OBJECTIVE:Gout nephropathy is a disease that causes damage to the kidney after gout caused by hyperuricemia.In recent years,with the deepening of research,high uric acid has caused the kidney's immune inflammatory damage to receive close attention.The rational use of China's rich traditional Chinese medicine resources is a new trend in treating the disease with good curative effect and less side effects.This experiment established a mouse model of gout kidney disease,observed the changes of renal function,renal pathology before and after treatment of Bi-ning Decoction,and detected inflammatory factors in kidney tissue,and preliminary explored the effect of Bi-ning Decoction on NLRP3 in mice with gout kidney disease model.METHODS:Male ICR mice,weighing 18-20 g,SPF grade,were adapted to the environment and were established gout kidney disease mice model by intraperitoneal injection of xanthine(0.6 g/kg)and potassium oxyoxalamate(0.3 g/kg)for 5 weeks.After the completion of modeling,high concentration group of Bi-ning Decoction,medium concentration group of Bi-ning Decoction,low-concentration group of Bi-ning Decoction,allopurinol group and saline group were intragastrically administered for 2 weeks.On the 14th day,the abdominal aorta was used to collect blood,and blood was taken to measure kidney function.The left kidney tissue was taken for paraffin embedding,and the right kidney cortex was used for protein detection.Statistical analysis was used to analyze the data of each group.Results:1.Changes in body weight:The weight of mice in the gout nephropathy model group was less than that in the control group,but it was not statistically significant(p>0.05).2.Serum indexes:Compared with allopurinol treatment group,the serum concentration of UA,Cr,and BUN in the high-concentration Bi-ning Decoction group was lower,and the difference was statistically significant(p<0.05).3.Changes of IL-1? detected by ELISA:Compared with the saline-treated group,the high-concentration treatment of Bi-ning Decoction significantly decreased the difference(p<0.01).4.Detection of NLRP3 changes by Western blot:Compared with saline-treated group,the expression of NLRP3 in inflammasome was significantly decreased in the treatment group and allopurinol treatment group,the difference was statistically significant(p<0.01);Compared with the purine alcohol group,the NLRP3 level in the high-medium concentration treatment group was significantly lower,and the difference was statistically significant(p<0.01).5.Kidney tissue was observed under light microscope:renal tubular degeneration and necrosis were most severe in the model group and the saline treatment group.A large number of inflammatory infiltrates of renal interstitium and scarring sclerosis were formed;tubular epithelial cell atrophy and degeneration in the allopurinol treatment group.Obviously,part of the interstitial fibrosis;high-concentration Bi-ning Decoction treatment group renal tubular epithelial cells atrophy,degeneration,renal interstitial lymphocyte infiltration reduced.Conclusion:Bi-ning Decoction high concentration group can significantly reduce the UA,Cr,BUN of mice with gout kidney disease,and reduce the expression of NLRP3 inflammasome and its inflammatory product IL-1?.The high-dose group of Bi-ning Decoction had certain protective and repairing effects on the kidneys of mice with gout-induced kidney disease.The allopurinol group had no significant improvement in NLRP3 in the kidneys of mice.
