| Objective: To investigate the expression levels of B7-H3 on peripheral blood mononuclear and inflammatory markers and miRNA-135 a in patients with habitual abortion,analyzing the changes of B7-H3 and inflammation markers in the course of habitual abortion,to provide the basis for exploring the pathogenesis of habitual abortion patients.Methods: 1.To collect 30 cases of normal pregnancy(NP),30 cases of history of abortion with pregnancy(HAP)and 50 cases of habitual abortion(HA)according to the principle of matching age and gestational age,after the normal pregnancy group collecting the specimens,follow-up data of the normal pregnancy group until delivery,to determine the normal pregnancy.2.B7-H3 on peripheral blood mononuclear cells was detected by flow cytometry,and serum soluble B7-H3(s B7-H3)was detected by enzyme linked immunosorbent assay(ELISA),Serum Th1/Th2 cytokines,including Th1 type: Interleukin 2(IL-2),Interferon-?(IFN-?)and Tumor necrosis factor-?(TNF-?),Th2 type: Interleukin 4(IL-4),Interleukin 6(IL-6),Interleukin 10(IL-10),were measured by the CBA method.3.Peripheral blood cell analysis was detected by hematology analyzer,and to calculate the neutrophil-lymphocyte ratio(NLR),platelet-lymphocyte ratio(PLR),and lymphocyte-monocyte ratio(LMR).4.Quantitative PCR was used to detect the level of serum miRNA-133 a and miRNA-135 a in patients with habitual abortion.Results: 1.The proportion of CD14+ monocytes in peripheral blood of HA group was lower than that of NP group and HAP group respectively(3.474±1.538% vs 4.857±1.613%,5.409±0.867%),with significant differences(P<0.05).The expression levels of B7-H3 on CD14+ monocytes in peripheral blood in HA group were significantly higher than those in NP group and HAP group(9.050±1.767% vs 1.327±0.641%,2.100±0.556%,respectively),with significant differences(P<0.05).There were no significant differences in the expression level of B7-H3 between the 3 months,6 months,and 9 months of gestational age of pregnant women.The serum s B7-H3 levels in HA group(2.700±0.130 ng/m L)and NP group(2.916±0.161 ng/m L)were higher than those in HAP group(2.369±0.172 ng/m L),there were significant difference(P<0.05).2.Levels of serum cytokines IL-2 in HAP group were higher than those in NP group,HA group(4.572±0.419 pg/m L vs 2.504±0.751 pg/m L?3.177±1.244 pg/m L),there were significant difference(P<0.05);Levels of serum cytokines IL-2 were also significantly higher in HA group than in NP group(P<0.05);Levels of serum cytokines IL-4 in HA group were significantly lower than those in NP group(2.730±1.666 pg/m L vs 3.500±1.395 pg/m L)(P<0.05).Levels of serum cytokines IL-6 in HAP group were higher than those in NP group an,HA group(3.808±1.153 pg/m L vs 2.689±1.004 pg/m L?2.357±1.048 pg/m L),there were significant difference(P<0.05);Levels of serum cytokines IL-10 in HAP group were higher than those in NP group,HA group(3.687±0.457 pg/m L vs 2.353±0.784 pg/m L?2.341±1.020 pg/m L),there were significant difference(P<0.05).3.The NLR in NP group was significantly higher than that in HAP group and HA group(3.877±1.250 vs 2.911±0.766,2.324±1.208),with significant difference(P<0.05);The PLR in HAP group was significantly higher than that in NP group(129.4±33.28 vs 104.4±33.41),with significant difference(P<0.05).The LMR in HA group was higher than that in NP group and HAP group(6.757±1.020 vs 3.780±1.262,4.807±1.485).The LMR in HAP group was higher than that in NP group,and there were significant differences(P<0.05).4.Levels of serum miRNA-133 a in HAP group was lower than those in NP group,there were significant differences(P<0.05);Levels of serum miRNA-135 a in HA group was significant lower than those in NP group,there were significant differences(P<0.05).Conclusions: 1.The levels of B7-H3 on peripheral blood mononuclear cells in patients with habitual abortion were highly expressed and there were imbalance of Th1/Th2 and bias Th1,it was suggested that B7-H3 may mediate the migration and activation of monocytes and then bind to the corresponding receptors on the surface of T cells and participate in the pathogenesis of habitual abortion.2.The rate of neutrophil-lymphocyte ratio(NLR)in patients with habitual abortion was low,it may be related to the secretion of a large number of inflammatory mediators from this type of patient's neutrophils to induce abnormal cell proliferation and accumulation of inflammatory cells,which in turn promotes the development of the course of the disease.3.The serum levels of miRNA-135 a in patients with recurrent miscarriage were low,which participate in the pathogenesis of habitual abortion. |
