Objective:By establishing the bacterial sepsis model of the wide-type infant mice,gal-3-/-infant mice,to test the bacterial removal rate of peripheral blood in two groups,to test the levels of IL-8?IL-10 and TNF-?,to compare the differences between the two groups of the phagocytic ability of macrophage and the maturity of phagosome,to analyze the role of gal-3 in sepsis.At the same time,the expression level of neonatal gal-3 in sepsis was detected to provide a new idea for the immune intervention of sepsis.Methods:1.Choosing 2-week-old wide-type infant mice and gal-3-/-infant mice,establishing the experimental sepsis model by intraperitoneal injection G-bacteria Salmonella typhimurium with 2.5x106CFU/one,no limit to male or female,dividing into two groups,n=20 in every group.2.Texting the expression levels of cytokines IL-8?IL-10 and TNF-? in macrophage of each group mice by ELISA.3.Comparing the bacterial clearance rate with the number of peripheral blood culture in trypsin soybean agar plate.By commonly incubating the isolated macrophage and Salmonella typhimurium to compare the intracellular bactericidal rate.4.Commonly incubating the peritoneal macrophages and carboxyfluorescein-SE labeled or carboxytetramethyrhodamine-SE labeled bacteria,to text average fluorescence intensity by the flow cytometry.Lyso-Tracker Red labeled the peritoneal macrophages,then commonly incubating with FITC labeled bacteria,Observing fluorescence reaction by The microscope.5.Testing the expression level of IL-8? TNF-??and gal-3 in normal and sepsis' s infant by ELISA.Results:1.Compared with the wide-type infant mice,the levels of IL-8 and TNF-? in the peritoneal macrophage of the gal-3-/-infant mice were not significant difference,but the level of IL-10 is low,and it has a statistical significance.2.Compared with the wide-type infant mice,the bacterial removal rate and the intracellular bactericidal rate of the gal-3-/-infant mice group was clearly inadequate.3.Compared with the wild-type infants,gla-3-/-mice phagocytic acidification was obviously deficient,the difference was statistically significant,and the phagocytic/lysosomal fusion was obviously deficient.4.Healthy term compared with healthy preterm,the expression levels of TNF-? and gal-3-/-were no significant difference,but the IL-8 was significantly higher.By comparing healthy term?pyemic term and healthy preterm?pyemic preterm,the expression of levers of IL-8?TNF-? and gal-3 increased obviously,the difference is statistically significant.Conclusion:1.Gal-3-/-mice were more susceptible to bacterial sepsis than those of the wild-type infants.2.Gal-3 mediated bacterial phagocytosis and phagocytic maturation in macrophage.3.Gal-3 participated in the inflammatory response in neonatal sepsis.
