| Objective:To observe the inhibitory effect of Dan Bei Yifu Granule on the proliferation of lung fibroblasts.and then observe the effect of all concentration of Dan Bei Yifu Granule on the expression of VEGF,VEGFR-1 and VEGFR-2 gene after the action of each time,thus confirming the effective therapeutic effect of Dan Bei Yifu in the anti pulmonary fibrosis.Then we will prepare the experiment to study the mechanism of the effect of Dan Bei Yi Fei Granule on pulmonary fibrosis.Methods:In the first stage,the lung fibroblasts were cultured with each concentration of danbei Yifu granule.Then 72h,48h and 24h were cultured at 5%C02 and 37 C.The effect of the cells was observed by microscope.After the action of each time,the lung fibroblasts were measured by MTT.The 490nm wavelength absorbance A value of the cell.In the second stage,the lung fibroblast culture was carried out with the concentration of Dan Bei Yifu granule,? 1mg/ml group of Dan Bei Yifu granule,? 1×10-1 mg/ml group of Dan Bei Yifu granule? 1 × 10-2 mg group of Dan Bei Yifu granule,? 1 × 10-3mg/ml group of Dan Bei Yifu granule,?Dexamethasone group.using immunohistochemical method and real-time quantitative PCR prescription The changes of VEGF,VEGFR-1 and VEGFR-2 expression in lung fibroblasts were measured.Results:The inhibitory rate of the concentration group(1×10-2 mg/ml)was close to the dexamethasone group(P>0.05),compared with the low dose group(1 × 10-3 mg/ml)(P<0.05)and the high dose group(1 × 10-1mg/ml)(P<0.05),which was more effective and had statistical significance.The interval is prolonged and rising to the highest 72h.The inhibitory rate of lung fibroblast proliferation was positively correlated with the time of the treatment of Dan Bei Yifei granules.Conclusion:The effect of Dan Bei Yifu Granule on pulmonary fibroblasts was effective,the expression of VEGF,VEGFR-1 and VEGFR-2 decreased,and the effect of anti pulmonary fibrosis was obvious,which showed obvious inhibitory effect. |
