Objective: This experiment applies the immunohistochemistry method to detect theexpression of VEGF-C and VEGFR-3in non-small cell lung cancer tissues, and tounderstand the relationship between the expression of VEGF-C, VEGFR-3and theclinicopathological characteristics of NSCLC. We study the correlation of VEGF-C andVEGFR-3in NSCLC tissues, and analyze the effect of VEGF-C and VEGFR-3on theprocess of NSCLC occurrence and development. We hope to provide a new referencemeans of NSCLC clinical diagnosis and treatment in the future.Methods: This experiment used immunohistochemistry method to detect the expressionsof VEGF-C (1:50) and VEGFR-3(1:80) in NSCLC group and the control group. In theexperiment all the NSCLC tissues were provided by the pathology department of YananUniversity Affiliated Hospital. All the specimens’ pathological diagnosis was confirmedby Hematoxylin–eosin dyeing. All the selected specimens had not been receivedchemoradiotherapy before the surgery. They were chose from August2005to January2012.We screened out65NSCLC tissue samples, which included52man patients and13female cases. Those specimens’ age was from38to74years old, and the average agewas61.50±10.50. According to WHO “classification standard”, those lung tissuesincluded49phosphorus carcinoma cases, and16adenocarcinoma cases. According to thehistological differentiation, they were grouped into two groups: high and middle degree53cases, low degree12cases. Moreover, there were34patients who had with lymphnode metastasis,31cases without lymph node metastasis. From the same period, weselected30adjacent cancer tissues and15benign lesions lung tissues as control group, which included5pulmonary inflammatory mass cases,3pulmonary bulla cases and7bronchiectasis cases. We applied the software SPSS17.0to analyze the date.Results:1.The expression rate of VEGF-C and VEGFR-3in NSCLC is higher than thosein adjacent cancer tissues and pulmonary benign lesions controls. P=0.000and P=0.003;2.VEGF-C’s expression in NSCLC groups: the expression rate of VEGF-C inphosphorus carcinoma、 adenocarcinoma cases is75.5%,56.3%, P=0.248,P>0.05;VEGF-C’s positive rate in the groups of age(≥50,<50years old) is75.6%,60.0%,P=0.203; VEGF-C’s positive rate in male and female groups is77.1%,52.9%, P=0.116;VEGF-C’s positive rate in the groups of high+middle and low differentiation is61.9%,87.0%, P=0.034, the difference is statistical significance.VEGF-C in the lowdifferentiated group expression is higher than it in high and middle tissues; the hyperexpressive rate of VEGF-C in lymph node metastasis is85.3%,which is more than54.8%which is VEGF-C’s expresses in no lymph node metastasis, P=0.007, the difference ismarkedly statistical significance;3.the expression of VEGFR-3in NSCLC groups: thepositive rate of VEGFR-3in phosphorus carcinoma, adenocarcinoma cases is71.4%,81.3%, P=0.654, the difference is not statistical significance; VEGFR-3’s positive rate inthe groups of age (≥50,<50years old) is75.6%,70.0%, P=0.638,P>0.05; VEGFR-3’spositive rate in male and female groups is78.2%,58.8%, P=0.187, P>0.05, it’s nostatistical significance; VEGFR-3’s positive rate in high and low differentiation degreegroups is64.3%,91.3%, P=0.018, the difference proves to be markedly statisticallysignificant. That result can show that VEGFR-3in low differentiated degree is hyperexpressive, while that expresses in high and middle tissues is lower than that in lowgroup.The positive rate of VEGFR-3in lymph node metastasis and no lymph nodemetastasis groups is85.3%,61.3%, P=0.028, the difference is statistical significance.The expression of VEGFR-3in no lymph node metastasis group is hyper expressive,however it is also lower than that in lymph node metastasis group.4. Using Spearmancorrelation analysis, we find there is a positive correlation between VEGF-C’s expressionand VEGFR-3’s expression (r=0.387, P=0.001). Conclusion:1.In non-small cell lung cancer tissues, VEGF-C and VEGFR-3expressover. Moreover, VEGF-C and VEGFR-3have a positive correlation2. The expressions ofVEGF-C and VEGFR-3in low differentiated degree lung cancer tissues are higher thanthose in high+middle differentiation degree tissues. Moreover, the expressions of them inthe lymph node metastasis group are higher than those in the group of no lymph nodemetastasis. VEGF-C and VEGFR-3take part in the process of NSCLC’s occurrence,development and lymph node metastasis.3. The expressions of VEGF-C, VEGFR-3haveno obvious correlation with tumor types, gender and age.
