ObjectiveSystemic lupus erythematosus?SLE?is a prototypic autoimmune disease,and lead to a variety of tissues and organs function damaged.Studies suggest that interleukin-35?IL-35?exerts protective roles in the pathogenesis and development of SLE.The aim of this study was to explore the associations between IL-35 gene polymorphisms and the susceptibility of SLE.MethodsA total of 490 SLE patients were recruited from Department of Rheumatism and Immunity,the First Affiliated Hospital of Anhui Medical University and Department of Rheumatology and Immunology,Anhui Provincial Hospital;and 489 healthy controls were recruited from Physical Examination Center of the First Affiliated Hospital of Anhui Medical University.Genotyping techniques(SNPscanTM)were used to explore the correlations between IL-35 rs2227314,rs2243115,rs2243123,rs2243131,rs428253,rs4740 and rs9807813 polymorphisms and the susceptibility and clinical manifestations of SLE.The plasma IL-35 levels were detected by ELISA in 76 SLE patients,the relationship between plasma IL-35 levels and IL-35 genotypes were analyzed.ResultsThere were no significant differences in the genotypes and allele frequencies of rs2227314,rs2243115,rs2243123,rs2243131,rs428253,rs4740 and rs9807813between SLE patients and healthy controls,respectively?all P>0.05?;under explicit model,implicit model and additive model,there were no significant differences in the genotypes of rs2227314,rs2243115,rs2243123,rs2243131,rs428253,rs4740 and rs9807813 between SLE patients and healthy controls,respectively?all P>0.05?.There were no significant differences in the genotypes and allele frequencies of rs2227314,rs2243115,rs2243123,rs2243131,rs428253 and rs9807813 between SLE patients with/without butterfly erythema,discoid rash,light sensitiveness,mouth ulcers,arthritis,serositis,kidney disease,nervous system abnormality,hematology abnormality and immunology abnormality,respectively?all P>0.05?;and there were no significant differences in the genotypes and allele frequencies of rs4740 between SLE patients with/without butterfly erythema,light sensitiveness,mouth ulcers,arthritis,serositis,kidney disease,nervous system abnormality and immunology abnormality,respectively?all P>0.05?,but there were significant differences in the genotypes of rs4740 between SLE patients with/without kidney disease and hematology abnormality,respectively;and there was a significant difference in the allele frequency of rs4740 between SLE patients with/without kidney disease.And there were no significant differences of plasma IL-35 levels of SLE patients in different genotypes of rs2227314,rs2243115,rs2243123,rs2243131,rs428253,rs4740 and rs9807813,respectively?all P>0.05?.ConclusionsThere were no significant correlations between the polymorphisms of rs2227314,rs2243115,rs2243123,rs2243131,rs428253 and rs9807813 of IL-35 and the susceptibility or clinical manifestation of SLE.And there were no significant correlations between the polymorphisms of rs4740 of IL-35 and the susceptibility of SLE,but there were significant correlations between the polymorphisms of rs4740 of IL-35 and the kidney disease and hematology abnormality of SLE,respectively,which suggested IL-35 gene polymorphism may be associated with the occurrence of certain clinical manifestations of SLE.
