Objective:To investigate the effects of parthenium lactone(PTL)on the proliferation and migration of human lung cancer A549 cells and its mechanism.Methods:Human lung cancer A549 cells were disposed with different concentrations of PTL,then the MTT assay was used to detected the proliferation ability of A549 cells;then cell migration ability was evaluated by scratch healing assay and Transwell migration assay;Flow cytometry was used to observe and detected the apoptosis of lung cancer A549 cells;apoptosis-related protein and EMT marker protein by Western blot.Results:1.The results of MTT assay showed that PTL significantly inhibited the proliferation of A549 cells in a time and dose dependent manner(P<0.05);2.The results of Flow cytometry showed that PTL could induce apoptosis of A549 cells(P<0.01);3.The results of scratch healing indicated that the migration ability of lung cancer A549 cells could be reducing by PTL(P<0.05);4.The results of Transwell invasion assay showed that PTL treatment significantly decreased the invasion ability of lung cancer A549 cells(P<0.05).5 Western blot results showed that the expression levels of Ezrin,Vimentin,Snail and Bcl-2 were significantly decreased(P<0.01),while the expressions of Cleaved-Caspases-3,Bax and E-cadherin were significantly increased in A549 cells treated with PTL(P<0.05 or p<0.01).Conclusion:1.The reduction of Ezrin expression may refer to the mechanism of PTL inhibiting proliferation and migration of lung cancer A549 cells;2.PTL can effectively reduce the proliferation,migration and EMT and induce apoptosis in A549 cells.
