Objective: To verify the effect of Maimendong Decoction on pulmonary fibrosis and to analyze its mechanism.Method: 1.Bleomycin-induced rat model of pulmonary fibrosis was divided into 4 groups according to different drugs: blank group: distilled water was administered by 1.1ml / 100g;model group: 24 h after modeling,Distilled water according to 1.1ml/100 g gavage;Maimendongtang group: 24 h after modeling,to Maimendongtang by crude drug 1.1g/ml/100 g gavage;prednisone group: 24 h after modeling,give strong Of the pine(prednisone acetate tablets)5mg / kg gavage.The effect of Maimendong Decoction on pulmonary inflammatory response in rats was detected by HE staining;the expression of collagen fibers in lung tissue was detected by Masson staining.2.Forty Sprague-Dawley rats(weight 200-220g)were randomly divided into 4 groups.The blank group was pretreated with 1.1ml/100 g distilled water,and the model group: After the model was established for 24 h,the distilled water was pretreated with 1.1ml/100 g.Maimendong Decoction group: 24 hours after the model was established,Maimendong Decoction was given by crude drug 1.1g/ml/100 g gavage;rh G-CSF(recombinant human granulocyte colony-stimulating factor)group: after modeling,10?g/(kg)· d)Dose subcutaneous injection of PBS-diluted rh G-CSF injection.The tail veins were collected from each group and the expression of CD44 and CD90 in serum of SD rats was detected by flow cytometry.3.GFP-BMSC cell lines,according to the different drugs,divided into DMEM basal culture medium group: rat BMSCs medium added the same amount of DMEM basal medium;Mai Mendong serum: rat BMSCs culture The serum containing 5% of Maimendong Decoction was added into the liquid;the serum of the model rats: the rat serum of BMSCs was added with 5% pulmonary fibrosis rat serum(Bleomycin group);the model rat Maimendong Decoction Serum group: 5% pulmonary fibrosis model serum and 5% Maimendong decoction serum were added to rat BMSCs culture fluid.The expression of SP-C,AQP-5 and their m RNA in each group of cells were detected by immunohistochemistry and q PCR,respectively.Result: 1.Compared with the control group,the degree of inflammation and the expression of collagen fibers in the Maimendongtang and prednisone groups were decreased;there was no significant difference between the Maimendongtang group and the prednisone group.2.Compared with the control group,the expression of CD44 and CD90 were up-regulated in the Maimendong Decoction group and the rh G-CSF(recombinant human granulocyte colony-stimulating factor)group;the Maimendong Decoction group and rh G-CSF(recombinant human granulocyte colony-stimulating factor)group,There were no significant differences between the groups.3.Compared with the control group,the expression of SP-C,AQP-5 and m RNA in the serum of the model rats of Maimendong Decoction was up-regulated.Conclusion: 1.Maimendong Decoction can reduce pulmonary fibrosis by reducing pulmonary fibrosis in rats with pulmonary fibrosis and reduce deposition of collagen fibers,thus achieving the purpose of treating pulmonary fibrosis;2.One of the mechanisms of Maimendong Decoction in treating pulmonary fibrosis is to promote the mobilization and differentiation of BMSCs in rat models of pulmonary fibrosis,that is,to increase the expression of CD44 and CD90 positive cells in lung fibrosis model rats.Its mobilization effect on BMSCs,followed by up-regulation of SP-C,AQP-5 and its m RNA expression levels demonstrated its differentiation effect on BMSCs.
