Background Nonalcoholic fatty liver disease(NAFLD)is becoming the most common chronic liver disease in the world.It is a range of related disorders including simple steatosis,steatohepatitis(NASH),liver fibrosis,cirrhosis and hepatocellular carcinoma.Several studies have showed that the process of NALFD has a strong link with the genetic and environment.Coronary heart disease(CAD)is one of the most common causes of death and the incidence is increasing recently in China.It is serious threat to human health.NAFLD and CAD have similar pathogenesis and risk factors include insulin resistance(IR),dyslipidemia,inflammation,oxidative stress,etc.These two diseases often exist in one patient.The genomic characteristics have been widely studied.The data from overseas population indicated that the polymorphism of MBOAT7 gene rs641738 is related to NAFLD and CAD.However,there is no research focus on the combined diseases and in Chinese Han population.Our study focus on the relationship between the MBOAT7 gene rs641738 and NAFLD with CAD in Chinese Han population.Objective In this study,we will investigate the association between polymorphism of PNPLA3 gene and CAD in Chinese Han Population in China.And to explore the possible mechanism of NAFLD with CAD,.MethodsThe study enroll 144 NAFLD patients,152 CAD patients,133 NAFLD with CAD patients and 140 healthy in Qingdao Municipal Hospital.Blood sample were obtained and collected for Genetic Analysis.Polymerase chain reaction analysis(PCR)and MatrixAssisted Laser Desorption/Ionization Time-of-Flight Mass Spectrometry(MALDI-TOF MS)were used for genetic analysis of MBOAT7 rs641738.The distribution of genotypes and alleles was determined by sequencing.The Hardy-weinbeurg was used to evaluate the representative of the group.At the same time,the general data,clinical data and laboratory data were collected.SPSS22.0 was used to analyze the data of genotypes and clinical features by chi square test,t test and Logsitic regression analysis.Results(1)There was no significant difference of the genotype of MBOAT7 rs641738 C/T polymorphisms the NAFLD group and heath controls(P=0.344).(2)There was no significant difference of genotypes and alleles between the CAD group,control group and NAFLD with CAD group(P= 0.873,P= 0.297,P=0.586).(3)In the overall,the carriers of CC and CT show a significantly lower serum cholesterol level(P = 0.036).(4)In the control group,the carriers of CT and TT had higher diastolic blood pressure and alkaline phosphatase than CC gene carriers(P=0.047?P=0.014).(5)In the NAFLD group,CT and TT genotype carriers had significantly higher ALT and TG(P=0.037,P=0.031).(6)In the CAD group,CT and TT genotype carriers had significantly lower aspartate aminotransferase and serum cholesterol lever than CC genotype carriers(P=0.029,P=0.013).(6)In the NAFLD group and the NAFLD with CAD group,the CT and TT carriers had lower diastolic blood pressure(P=0.002).Conclusion MBOAT7 rs641738 polymorphism may not be an independent risk factor for NAFLD,CAD,NAFLD with in China.
