Association Between Polymorphisms Of FABP1, FABP2 Gene And Nonalcoholic Fatty Liver Disease In Uygur Population Of Xinjiang Region

Objective: The aim of the present study was to investigate the association of Liver Fatty acid binding protein(L-FABP,FABP2) and Intestinal fatty acid-binding protein(I-FABP,FABP1) gene polymorphisms with NAFLD in a Uyghur population of Xinjiang. Methods: In this case-control study,304 cases of NAFLD patients were recruited in NAFLD group and 304 healthy examination individuals were selected as control. Clinical data and blood sample was collected from each subject, genomic DNA was extracted from whole peripheral blood leukocytes. The FABP1,FABP2 gene polymorphism was detected by improved multiple ligase detection reaction(i MLDR). Analyse the relationship between susceptibility to NAFLD and different genotypes. All statistical analysis was performed by using SPSS 17.0 software. Results:1.The levels of BMI, Waist circumference, DBP、SBP、 TC、TG、LDL-C、UA、AST、ALT、FPG were significantly higher than that of control group, but HDL-C are lower than the control group(P<0.05)。2.The familial history of hypertension and diabetes were significantly higher than that of control group,(P<0.05), and There was no difference between the two groups in the family history of tumor and coronary heart disease.3.Multivariate logistic regression analysis showed that the fresh fruits, fresh vegetables, physical exercise is a protective factor of NAFLD, and eating meat, snack food, drinking is a risk factor of NAFLD. in addition, obesity, central obesity, high uric acid, low density lipoprotein cholesterol, high density lipoprotein were increase the risk of NAFLD.4.The frequencies of GG, GA and AA genotype on FABP1 rs1545224 in NAFLD group were 6.2%、43.5% and 50.3%, respectively; where as those in control group were 5.50% 、 34.75% and 59.75%, respectively. Significant differences in the frequencies of FABP1 rs1545224 genotype were found between NAFLD and control group(χ2=13.02,P=0.001). The frequencies of G and A allele on FABP1 rs1545224 in NAFLD group were 27.9% and 72.1%, respectively;where as those in control group were 22.88% and 77.12% respectively. Significant differences in the frequencies of FABP1 rs1545224 allele were found between NAFLD and control group(χ2=9.614,P=0.020). Univariate Logistic regression analysis showed that, compared with the G allele population, carrying A alleles can decrease the risk of NAFLD.(G vs A,OR=0.766,95%CI 0.647~0.907). There is no Significant differences in the frequencies of CC、CT、TT genotype and C、T allele on FABP1 rs2241883 and FABP2 rs179988 in NAFLD group(P>0.05). 5.in all research objects a significant association between rs1545224 and NAFLD risk could also be found in additive genetic model(GG + GA vs AA, P=0.000, OR=0.683 95%CI=0.553~0.842) and in female subjects a significant association between rs1545224 and NAFLD risk could also be found in additive genetic model(GG+GA vs AA,P=0.001,OR=0.621,95%CI=0.467~0.825).6.in patients with NAFLD, SCr levels between rs1545224 genotype GG genotype > AA genotype >GA genotype, DBP level for GA genotype > GG genotype > AA genotype and uric acid level of AA genotype > GG genotype >GA genotype; in patients with NAFLD and ALT among different gene types in FABP1 locus gene rs2241883, AST level for CC genotype >TT genotype >CT genotype, DBP level for CC genotype >CT genotype >TT genotype. Uric acid level was >CC genotype>TT genotypeCT genotype.Conclusion:1.obesity,abdominal obesity,high uric acid and low HDL levels,high and low density lipoprotein were risk factors of nonalcoholic.2.The polymorphisms of rs1545224 in NAFLD gene may be associated with NAFLD in Uyghur population from Xinjiang region, carrying A alleles can decrease the risk of NAFLD, there is no siginificant association between FABP1 gene rs2241883 and rs1799883 gene FABP2(Ala54Thr) with non alcoholic fatty liver disease in Uyghur population in Xinjiang region.3.The rs1545224 polymorphism of FABP1 gene has influence on level of Cr、DBP、BMI、UA and FBG level; The rs2241883 polymorphism of FABP1 gene has influence on level of BMI 、waist circumference、TG、TC、DBP、AST、ALT 、UA;The rs1799883 polymorphism of FABP2 gene has influence on level of BMI and glycosylated hemoglobin.
