Purpose: Preeclampsia is a pregnancy disorder characterized by the first hypertension and proteinuria after 20 weeks of gestation.It is a major cause of maternal and fetal maternal and fetal death in the world.So far,the pathogenesis of the disease has not been fully explained.In this paper,we used homocysteine to induce preeclampsia and eclampsia disease model in pregnant mice to detect the expression of m Glu R5 m RNA and protein in hippocampus of rats in each group.It is speculated that it may play a possible role in the occurrence and development of pre-eclampsia and eclampsia,and providing new idea for exploring the pathogenesis of preeclampsia and eclampsia in humans.method:1.Model establishment: According to complete randomization method,the healthy mature adult Wistar female and male rats were selected and female and male rats were housed in the same cage at a ratio of 4:1.The vaginal secretions were tested daily,and the pregnancy was determined after the sperm was detected.It was defined as the 0th day of pregnancy.The selected rats were randomly divided into 4 groups:non-pregnancy control group(NN),non-pregnant homocysteine group(NH),pregnancy control group(PN),and pregnancy homocysteine group(PH).10 rats in every group.From the 10 th day of gestation,NN group and PN group were intraperitoneally injected with physiological saline,and NH group and PH group were intraperitoneally injected with homocysteine(Hcy).2.Detection index: after treatment,the caudal artery systolic pressure,24-hour urinary protein content and biological behavior of each group were monitored.On the 20 th day,blood gas analysis was performed after anesthesia.The number of fetal rats was recorded after cesarean section.Fetal weight.At the same time,hippocampal tissue was isolated from rats after anesthesia,RNA in rat hippocampus tissue was extracted,and then reverse transcribed into c DNA.Real time PCR and Western blot were used to detect the expression of m Glu R5 m RNA in hippocampus.result:1.Blood pressure: There was no significant difference in basal blood pressurebetween the groups.Blood pressure in the PH and NH groups increased fromthe 12 th day.The increase in blood pressure in the PH group was greater thanthat in the NH group during the first half of the administration period.Thedifference was significant(P<0.05).The increase in blood pressure was similarin both groups,but there was a significant difference compared with the othertwo groups(P<0.01).2.Urinary protein: There was no significant difference in basic urinary proteinbetween the groups.The urinary protein levels in the 24 hours of the PH groupwere significantly higher than those in the other 3 groups on the 15 th and 19 thdays of pregnancy(P<0.01).There was no significant difference between thethree groups(P>0.05).3.Biological behaviors: There were different degrees of behavioral changes inthe PH group during late pregnancy,such as decreased activity,staring,andsluggishness.Morris water maze test showed a prolonged total time and totaldistance,compared with the other three groups,the difference was significant(P<0.01).4.Rats' weight: The fetal weight of the PH group was significantly lower thanthat of the PN group(P<0.01),5 stillbirths,and the stillbirth rate was 4.8%.5.Blood gas analysis: There was a tendency of metabolic acidosis in differentdegrees in all four groups,but the most serious was in PH group,and thedifference was significant(P<0.01).6.Real-time PCR and Western blot were used to detect the expression of m Glu R5:the expression of m Glu R5 in hippocampus of pregnant rats in PH group wassignificantly increased compared with other three groups,and the differencewas statistically significant(P<0.01).Conclusion: The increased expression of m Glu R5 m RNA and protein in brain tissue of pregnant rats may had a positive effect in the occurrence of eclampsia..
