Objective: To evaluate the effect of warm acupuncture on lipid metobolism through several indexes as blood lipids,blood rheology and ATP binding cassette transporter A1(ABCA1),liver X receptor ?(LXR?),Interleukin 1?(IL-1?)and Tumor necrosis factor-?(TNF-?)of experimental hyperlipidemia rats in order to explore its mechanism in prevention and cure hyperlipidemia lesions.Methods: 40 healthy male SD rats were randomly divided into four groups,the normal controlgroup A,the model group B,the warm acupuncture group C,the acupuncture group D,10 rats in each group.After a week of adaptive feeding,except for the normal group,the rats in other groups were fed with high fat diet for 4 weeks to establish experimental hyperlipidemia model.At the end of the model period,the serum samples were collected from the rats by the method of bleeding tails,and the Serum TG,TC level were analyzed to evaluate the model is successful or not.After successful modeling,all rats were fed continuously according to the original plan,meanwhile group C was given warm acupuncture to intervene,group D was given acupuncture to intervene.During the experiment,the weight,hair color,activity and mental status of the rats in each group were observed carefully.After the treatment,blood,liver and aortic arch tissue were collected.The level of TG,TC,LDL-C, HDL-C,blood rheology and expression of ATP binding cassette transporter A1(ABCA1),liver X receptor?(LXR?),IL-1? and TNF-? were measured.Result: The general conditions of the rats:There were no deaths in all rats during the experiment.The rats in the normal control group were in good general condition during the experiment.The rats in other groups gradually appeared the phenomenon of poor spirit,decreased activity,dry hair.Compared with the normal control group,the hair color and activity of the rats in the treatment group were slightly worse,but better than those in the model group.1.Comparison of lipid levels: The serum TC,LDL-C level of the model group were higher while the HDL-C were obsviously lowert than that of the normal group(P<0.01,P<0.05).Compared with the model group,TC,LDL-C of warm acupuncture group and acupuncture group were significantly decreased,while HDL-C levels were significantly increased(P<0.05).The TC in warm acupuncture group were lower than that of acupuncture group(P<0.05).2.Comparison of hemorheological indexes:The whole blood viscosity,plasma viscosity,HCT of the model group was significantly higher than that of the normal group.Compared with the model group,those four indexes in warm acupuncture group and acupuncture group decreased statistical significance(P<0.05).3.Comparison of ABCA1 and LXR? in liver and aortic arch of rats in each group:Compared with the normal group,the level of expression of ABCA1,LXR? m RNA were significantly decreased in the model group(P <0.05);ABCA1 and LXR? m RNA in the warm acupuncture group and acupuncture group were significantly increased than those of the model group.(P<0.05);compared with the acupuncture group,those two indexes in the warm acupuncture group were significantly increased(P <0.05).4.Comparison of protein levels of IL-1? and TNF-? in liver tisse:As compared with the normal group,IL-1? and TNF-? levels were higher in the model group(P<0.05).As compared with the model group,IL-1? and TNF-? were significantly decreased in the warm acupuncture group and acupuncture group(P<0.05).As compared with the acupuncture group,those two indexes were significantly decreased in the warm acupuncture group(P <0.05).Conclusion: This study indicated that warm acupuncture at Fenglong and Zusanli could obviously down-regulate the levels of TC and LDL-C in hyperlipidemia rats,improve hemorheological indexes,promote the expression of ABCA1 and LXR? in liver and aortic arch tissue,and to inhibit the expression of IL-1? and TNF-?,thus promoting the process of reverse cholesterol transport and intervene the AS caused by Hyperlipidemia effectively.
