Objective:The purpose of this study was to investigate the association between KLF14 gene polymorphism and coronary heart disease in Han nationality in Qingdao by detecting genetic polymorphisms of CHD patients and healthy people in Han nationality in Qingdao.Participants and methods:The collection of Qingdao Municipal Hospital Department of cardiology from March 2016 to November 2017 for diagnosis and treatment of patients,including 241 cases of coronary heart disease patients,116 cases in the control group,coronary heart disease patients after PTCA(coronary angiography)diagnosis of coronary artery stenosis,while recording the degree of disease and vascular lesions;the control group after coronary angiography confirmed vascular smooth or the coronary artery stenosis CTA.The subjects of two groups were BMI,age,gender,smoking history,drinking history,history of hypertension,fasting blood glucose,blood lipids,diabetes history data were recorded.Extended primer PCR reaction first design and synthesis of target gene and single gene by biotechnology company synthesis,extraction and then save spare DNA,two polymorphic loci rs972283,rs4731702 and Agena using Mass Array DNA mass spectrometry to detect KLF14 gene system gene and genotype distribution of KLF14 gene,allele frequency and the genotype frequency in the two groups,and the clinical data between different genotypes were analyzed.At the same time,the gene frequency in patients with coronary heart disease coronary lesions and genotype frequencies were analyzed and compared.Data processing using SPSS 19 software.The study population balance test samples for Hardy Weinberg polymorphic loci genotypes,analysis variables were used-test,the main observation indicators were normality test,if it belongs to the skewed distribution of data,through the relevant conversion method can get the approximate teenage boy data distribution;continuous variable data using t test analysis.P<0.05 had significant difference.Result:1.The clinical data of coronary heart disease group and control group were compared between FPG,TC,TG,HDL-C,LDL-C,Protein)in coronary heart disease group and control group no significant difference,no statistical significance(P>0.05);2.The group of coronary heart disease in smoking,drinking,diabetes,male,BMI was significantly more than the control group(P<0.01)3.Rs972283 site coronary heart disease group and control group KLF14 gene AA,AG,GG frequency is respectively 7.1%,42.7%,50.1%,1.4%,34.3%,65.3%,the CHD group rs972283 A allele frequency(27.8%)and control group(26.5%)of similar frequency(P>0.05),CHD group and control group the gene mutation frequency has no obvious difference,no statistically significant difference(P>0.05);4.coronary heart disease group and control group KLF14 gene rs4731702 genotype CC,TT and CT frequency were 49.8%,7.1%,33.1%,56.7%,5.2%,38.1%,the CHD group rs4731702 T allele frequency(28.6%)and control group(24.8%)of similar frequency(P>0.05),between CHD group and control group the mutation frequencies had no significant difference,no statistically significant difference(P>0.05);5.KLF14 gene polymorphism in coronary heart disease groups of different coronary lesions group no significant difference.6.There were significant differences in KLF14 gene loci rs972283 in CHD group in the diabetic group(P<0.05),no significant difference of rs4731702 in CHD group in the diabetic group(P>0.05)7.There was no significant difference in 7..KLF14 gene loci rs972283 and rs4731702 in coronary heart disease group in hypertension group(P>0.05)Conclusion:1.No significant correlation between KLF14 gene polymorphism and coronary heart disease in the Han population in Qingdao area;2.KLF14 gene polymorphism in coronary heart disease group of different coronary artery disease in subgroups had no significant difference,there is no obvious correlation3.No significant correlation was found between.KLF14 gene rs4731702 polymorphism and coronary heart disease in the Han population in Qingdao area with diabetes,there is a significant correlation between rs972283 and Qingdao Han population of coronary heart disease and diabetes4.There was no significant correlation between hypertension with coronary heart disease in the Han population KLF14 gene loci rs972283 and rs4731702 in Qingdao area...
