Objective Detection of tumor necrosis factor-alpha(TNF-?),interferon-gamma(IFN-?)and interleukin-17(IL-17)in patients with spinal tuberculosis The expression of TNF-?,IFN-? and IL-17 in the lesions and their expression in serum were used to investigate the correlation between the expression of TNF-?,IFN-? and IL-17 in the lesions and the expression level in the serum.Methods From June 2016 to February 2018,patients with spinal tuberculosis who received surgery for orthopaedic surgery in the People's Hospital of Guangxi Zhuang Autonomous Region were recruited.According to the inclusion and exclusion criteria,15 patients were included in the study,including 9 males and 6 females.Between the ages of 24-80 years old.Blood samples were collected at the time of admission and about 2 weeks after systemic antispasmodic treatment,selective removal of spinal tuberculosis and titanium cage grafting and internal fixation were performed.Spinal tuberculosis lesions were obtained during surgery.Enzyme-linked immunosorbent assay(ELISA)was used to detect the expression of TNF-?,IFN-? and IL-17 in serum and tuberculosis lesions,respectively.The t-test and Pearson's method were used to analyze the correlation of the data.Results(1)The relative expression level of TNF-? in serum was 281.84 37.31 pg/ml and 871.60 74.12 pg/ml in lesions;the relative expression level of IFN-? in serum was 56.09 14.78 pg/ml,and the lesion was 39.50. 10.38pg/ml;relative expression of IL-17 in serum was 60.23 11.00 pg/ml,and 70.80 13.07 pg/ml in lesions.The difference in the relative expression of these three factors in serum and lesions was statistically significant(P<0.01).In the similarity search,the relative expression levels of TNF-?,IFN-?,and IL-17 in the control group(non-spinal tuberculosis group)were approximately 200pg/ml[1],25 pg/m[2]l,and 35 pg/ml[3],respectively.(2)There was a significant positive correlation between the relative expression levels of TNF-?,IFN-? and IL-17 in serum and in lesions(TNF-alpha correlation coefficient r=0.567,P<0.05,IFN-gamma correlation coefficient r=0.876,P<0.01,IL-17 correlation coefficient r = 0.929,P < 0.01)(3)There was a significant positive correlation between serum TNF-? levels and IL-17 levels(correlation coefficient r=0.672,P <0.01);There was also a significant positive correlation between TNF-? levels and IL-17 levels in the lesions(correlation coefficient r = 0.533,P<0.05).Conclusion(1)To find the expression levels of TNF-?,IFN-? and IL-17 in the control group(non-spinal tuberculosis patients)in a large number of similar trials,the TNF-?,IFN-? and IL-17 levels in serum and The relative expression levels in the lesions were higher than those in the reference group,suggesting that TNF-?,IFN-? and IL-17 may play an important role in the development and evolution of spinal tuberculosis.This may have some reference value for the early diagnosis of spinal tuberculosis.(2)There was a significant positive correlation between TNF-?,IFN-? and IL-17 levels in serum and lesions in patients with spinal tuberculosis,suggesting that serum TNF-?,IFN-? and IL-17 levels may be expressed.Levels to infer immune expression of relevant factors in tuberculosis lesions.(3)There was a significant positive correlation between serum TNF-? levels and IL-17 levels and TNF-? levels in the patients with spinal tuberculosis and IL-17 levels,indicating that TNF-? may be an important factor in regulating IL-17 expression in spinal tuberculosis.one.
