Purpose and significance:To investigate the regulation of the glucagon like peptide-1 receptor agonist lllagin on the inflammatory response of human umbilical vein endothelial cells(HUVECs)induced by PCI and the related molecular mechanism.Method:Resuscitate and culture cryopreserved human umbilical vein endothelial cells,take 3-5 generations of cells for experiments,use sera from patients with post-PCI to induce HUVECs inflammatory lesions to establish endothelial inflammatory response models and set negative control groups,then give liraglutide for Interventions and groupings for research.Mtt assay was used to detect the activity of human umbilical vein endothelial cells with different concentrations of liraglutide,then calculate the proliferation rate of cells in each group and measure LDH leakage rate.Nitric oxide(NO)content in each cell was determined by nitrate reductase method and NOS activity was determined by colorimetry.The contents of cytokines MCP-1,sVCAM-1,sICAM-1,IL-1 and IL-6 in each group were determined by ELISA.RT-qPCR was used to detect the expression of Bcl-2 and Bax genes related to apoptosis.Western-blot detected PKA and NF-?B-p65 activation levels.Results:The expressions of cytokines MCP-1,VCAM-1,ICAM-1,IL-1 and IL-6 in serum group after PCI were higher than those in negative control group(p < 0.05).compared with serum group after PCI,MCP-1,VACM-1,ICAM-1,IL-1 and IL-6,the expressions of apoptosis factors bcl-2 and bax were significantly decreased in liraglutide treatment group(p < 0.05).No content and nos activity in serum group after PCI were significantly lower than those in negative control group(p < 0.05),and that in liraglutide treatment group was significantly higher than that in serum group after PCI(p < 0.05).LDH leakage rate in liraglutide treatment group was significantly lower than that in serum group after PCI(p < 0.01).The activation level of NF-?b protein in serum group of patients after PCI increased significantly,while that of PKA protein decreased significantly.Liraglutide can inhibit the activation level of NF-?B-p65 and promote the activation level of PKA.Conclusions:Liraglutide has protective effect on inflammatory damage of human umbilical vein endothelial cells in serum after PCI,and its effect may be mediated by PKA /NF-?B signaling pathway.
