Objective:To explore mechanism of peripherally inserted central catheter(PICC)indwelling and injury of 5-FU stimuli on human umbilical vein endothelial cells(EA.HY926).Methods:Firstly,sterile PICC catheter was used and tailored into proper length to put into EA.HY926 cell culture fluid.Add 5-FU to make a 40ug/mL concentration for joint culture of EA.HY926 cells.Model of PICC indwelling and human umbilical vein endothelial cells injury induced by 5-FU would be set up.Secondly,MTT method was taken to examine activity change of EA.HY926 cells at different time intervals in control group and each model group(PICC injury group,5-FU injury group and PICC-combined 5-FU injury group).Next,scratch test was adopted to detect change of EA.HY926 cells mobility at different time intervals in control group and each model group.Then,ELISA method would be taken to examine expression level of related inflammatory factors vWF,IL-6 and FIB in EA.HY926 cell culture supernatant fluid in control group and each model group.Finally,Western-Blotting was taken to check expression level of vWF,IL-6 and FIB protein within EA.HY926 cells in control group and each model group.Results:1.Compared with control group,cell activity after being cultured for 30 min,60min and 120 min in each model group was reduced and the difference was of statistical significance(P<0.05),among which cell activity in mimicked PICC combined 5-FU injury group was the lowest.2.Comparison of mobility in each group.Mobility in PICC injury group,5-FU injury group,PICC combined 5-FU injury group and control group was 51.72%,21.57%,18.24% and 75.94% respectively.Mobility in each model group was decreased compared with that in control group and the difference was of statistical significance(P<0.05).Among,mobility in mimicked PICC-combined5-FU injury group was the lowest.3.When compared with control group at 30 min of induction,vWF concentration in cell culture supernate in each model group was all enhanced and the difference was of statistical significance(P<0.05).Concentration of IL-6and FIB at 30 min,60min and 120 min of induction in each model group was increased compared with that in control group and the difference was of statistical significance(P<0.05).Among,IL-6 and FIB concentration elevation was most obvious in mimicked PICC-combined 5-FU injury group.4.After induced for 24 h,expression level of vWF,IL-6 and FIB protein in each model group were improved compared with that in control group and the difference was of statistical significance(P<0.05).Among,expression level of vWF,IL-6 and FIB protein in mimicked PICC combined 5-FU injury group was the most obvious.Conclusion:PICC indwelling combined 5-FU would damage human umbilical vein endothelial cells,reduce cell activity,inhibit cell mobility and cause cell injury through releasing vWF.Change of inflammatory factors IL-6 and blood coagulation factor FIB concentration can reflect to a certain degree the inflammation reaction after EA.HY926 cell injury caused by PICC indwelling and 5-FU stimuli,offering reference for future exploration of the mechanism of vascular injury caused by PICC indwelling combined 5-FU.
