| Part I Epidemiological characteristic of HIV/AIDS in a HIV confirmation laboratory in Guangxi,2014-2016Objective: Analysis on epidemiological characteristics and trends of HIV/AIDS in HIV confirmation laboratory of a general hospital in recent three years,and provides the disease epidemic information for the further formulation and implementation of AIDS prevention and control policy.Method: Analysis of HIV/AIDS epidemic characteristics from 2014 to2016 in HIV confirmation laboratory of the First Affiliated Hospital of Guangxi Medical University by descriptive epidemiology.Results: From 2014 to 2016 a total of 1412 HIV/AIDS patients were diagnosed in laboratory,including 1119 male cases and 293 female(male to female ratio of 3.8: 1).Compared with the same period in three years,the number of AIDS detection has increased.The HIV/AIDS has a wide epidemic area,the floating population is the main infected people and only 40.44% inNanning;The majority of the infected people were farmers,accounting for48.08%;Youth category was high risk group,accounting for 41.01%;Most of cases were junior high school or lower education level.In this study,the proportion of male to female with HIV/AIDS patients was higher than that of AIDS in Guangxi,and the spatial distribution of HIV/AIDS patients was also different.However,high-risk groups were mainly youth category and middle-aged,junior high school or lower education level,rural residents /migrant workers.Conclusion: In the past three years,the number of AIDS infections has been greatly increased,the epidemiological characteristics of the disease are very different,and male as the main infected population,youth category and middle-aged group,low-level education for high-risk population.Effective prevention and control measures should be adopted for different groups to reduce the new cases.Part II Expression and Function of WNT Gene in HIV-1 Infected PatientsObjective: To study the differential expression of Wnt genes in HIV-1infected patients,and analyzed by sex,age,CD4+T cell counts and viral load.The differential expression of Wnt protein in plasma was tested by ELISA.And preliminary analysis of the role of Wnt gene in HIV-1/AIDS,to provide the basis for the study of HIV-1/AIDS mechanism,AIDS vaccine and treatment.Method: 28 HIV-1 infection patients and 33 healthy individuals were brought into this study,the relative expression of Wnt genes in peripheral blood were detected by Real-time PCR,and the sex,age,CD4+T cell counts and viral load were collected.The comparison of the relative expression levels of Wnt genes between the two groups was compared by using the Wilcoxon rank sum test,and the stratification analysis and correlation analysis were carried out according to sex,age,CD4+T cell counts and viral load,respectively.The concentrations of Wnt1 and Wnt3 in plasma were validated by ELISA.Results: 1.The average age of HIV-1 infection patients was 44.11 ± 16.06 years old,including 23 males and 5 females,with predominantly Zhuang and Han,and CD4+T cells were 12-642 cells/μL;the viral load was 34.3-818,000copies/m L.2.There was no significant difference in Wnt 8a m RNA between the two groups(P>0.05).The expression of Wnt2,Wnt3,Wnt5 a and Wnt8 b in HIV-1infected patients was higher than those in healthy individuals(P<0.05);The expression of Wnt1、Wnt2b、Wnt3a、Wnt4、Wnt5b、Wnt6、Wnt7a、Wnt7b、Wnt9a、Wnt9b、Wnt10a、Wnt10b、Wnt11、Wnt16 in HIV-1 infection were lower than those in healthy individuals(P<0.05).The concentration of Wnt1 in patients with HIV-1 infection and healthy individuals were 1.00±0.14 ng/m L、1.09±0.12 ng/m L,respectively;and the concentration of Wnt3 were 0.43±0.18ng/m L、0.31±0.14 ng/m L,respectively.The concentration of Wnt1 protein in HIV-1 infected patients is lower than healthy individuals,and the concentration of Wnt3 protein is higher than healthy individuals,consistent with the results of Real-time PCR.3.The sex and age in HIV-1 infected patients had no effect on Wnt genes expression.4.According to CD4+T cell count and viral load,the relative expression of Wnt7 a m RNA was decreased in patients with CD4+T cells <200 cells /μL(Z=-2.091,P=0.037);There was no significant difference in the expression of Wnt genes in patients with virus load >20 copies / m L or < 20 copies / m L(P>0.05).Spearman rank correlation analysis showed that the expression of Wnt7 a m RNA was positively correlated with the number of CD4+T cells(r=0.417,P=0.027).The expression of Wnt7 a m RNA and Wnt10 a m RNA was negatively correlated with viral load(r Wnt7 a m RNA=-0.519,PWnt7 a m RNA=0.005;r Wnt10 a m RNA=-0.428,PWnt10 a m RNA=0.023).Conclusion: 1.The expression of Wnt genes in HIV-1 infection patients was significantly different from those in healthy individuals.Some genes were up-regulated and most of them were down-regulated.Suggesting that after infected with HIV-1 the Wnt signaling pathway abnormal activation or inhibition.2.The expression of Wnt7 a m RNA was positively correlated with the number of CD4+T cells,it is suggested that Wnt7 a may be used to mediate the expression of CD4-CD8+ on the cell surface,and indirectly reflect CD4+T cells and reflect the immune function.3.Wnt7 a and Wnt10 a may act as signaling molecules that inhibit the replication of HIV-1 in PBMC,which may serve as a target gene in future studies,provide a basis for the study of HIV-1 treatment. |
