Objective: To observe and compare the effects of etomidate or propofol on early spatial cognition,spatial exploration,learning and memory abilities,plasma S100 ? protein,partial oxygen pressure and hippocampus neurons in young rats.Methods : one hundred and thirty-five SD rats(31 ~ 40 g)aged 21 ~ 22 days were randomly divided into 9 groups which were screened by Y-maze,15 young rats in each group.According to the anesthetic and oxygen concentration inhalation divided group air-control(CO): 10ml/kg of normal saline was intraperitoneal injected singly and expose air 2 hours,group middle concentration oxygen-control(MO):10ml/kg of normal saline was intraperitoneal injected singly and expose 45%~50% concentration of oxygen 2 hours,group high concentration oxygen-control(HO): 10ml/kg of normal saline was intraperitoneal injected singly and expose 95%~100% concentration of oxygen 2 hours,group air-etomidate(CE): 5ml/kg of etomidate was intraperitoneal injected singly and expose air 2 hours,group middle concentration oxygen-etomidate(ME): 5ml/kg of etomidate was intraperitoneal injected singly and expose 45%~50% concentration of oxygen 2 hours,group high concentration oxygen-etomidate(HE): 5ml/kg of etomidate was intraperitoneal injected singly and expose 95%~100% concentration of oxygen 2 hours,group air-propofol(CP): 50ml/kg of propofol was intraperitoneal injected singly and expose air 2 hours,group middle concentration oxygen-propofol(MP): 50ml/kg of propofol was intraperitoneal injected singly and expose 45%~50% concentration of oxygen 2 hours,group high concentration oxygen-propofol(HP): 50ml/kg of propofol was intraperitoneal injected singly and expose 95%~100% concentration of oxygen 2 hours.Etomidate or propofol anesthesia awake and oxygen inhalation after 2 hours,the test of OFT,HBT and Y-maze were perfomed(n=5).Test the content of S100? protein which was collected by plasma of inferior vena cava(n=5).Blood samples were collected from abdominal aorta and blood gas analysis was performed to detect oxygen partial pressure.Observe the morphology of hippocampus neuron tissue by HE staining.Results:(1)In the OFT test,there were no significant difference between group CO?MO?HO?CE?ME?HE?CP?MP and HP about the locomotion(P>0.05).(2)In the HBT,compared with group CO,group MO about times of young rats stretch into the hole were increased but no significant difference(P>0.05),HO?CE?ME?HE?CP?MP?HP were all decreased(P<0.05);there were no significant difference between group CE,ME and HE(P>0.05);there were no significant difference between group CP,MP and HP(P>0.05);compared with group MO,group ME,MP were all decreased(P<0.05);compared with group HO,group HE,HP were all no significant difference(P>0.05).(3)In the Y-maze test,there were no significant difference between group CO,MO,HO,CE,ME,HE,CP,MP and HP about RN and TRT.(4)The result of oxygen partial pressure,there were no significant difference between group CO,CE and CP(P>0.05);there were no significant difference between group MO,ME and MP(P>0.05);there were no significant difference between group HO,HE and HP(P>0.05);compared with group CO,CE and CP,group MO,ME,MP,HO,HE and HP all increased(P<0.05);compared with group MO,ME and MP,group HO,HE and HP all increased(P<0.05).(5)The S100? content test by ELISA,compared with group CO,group MO,HO,CE,ME,HE have no significant difference(P>0.05),whereas group CP,MP,HP were all increased(P<0.05);there no significant difference between group CP,MP,HP(P>0.05).(6)The morphology of hippocampus neuron tissue by HE staining.The neural cells in the brains of the nine groups were uniform and dense,arranged regularly and orderly.The hippocampal neurons had complete structure,clear nucleoli,abundant cytoplasm and no obvious degeneration and necrosis of cells.Conclusions:(1)Exposed to 95%~100% concentration of oxygen two hours can make effects the young rat' ability of exploration,but no influence of the spatial cognition and learning and memory.(2)Both etomidate and propofol have potential cognitive protection and injury effects on young rat' ability of exploration exposed to 95 % ~ 100 % oxygen for 2 hours.(3)Compared with the air exposure environment,etomidate and propofol had no further space exploration ability damage under 45 % ~ 50 % medium oxygen and 95 % ~ 100 % high oxygen.(4)A single propofol dose may lead to an increase in s100? protein in the plasma of young rats;different concentrations of oxygen exposure and combined etomidate or propofol had no significant effect on hippocampal tissue in young rats.
