Objective:An intestinal ischemia reperfusion (I/R) rat model was adopted by blocking the SMA to observe the morphologic change of retina and the influence of index that correlated with oxygen free radical (OFR) damage, then the influence of propofol and etomidate on above-mentioned correlated index. The aim of this study was to evaluate the protective effect of propofol and etomidate on retinal injury, and its possible mechanisms, in order to provide some empirical study information for illuminating mechanisms and basic reference data for its clinical application.Methods:32 male and healthy Wistar rats were randomly divided into 4 groups as follows:sham operation group (Group S), intestinal ischemia reperfusion group (Group I), propofol group (Group P) and etomidate group (Group E). Each group accepted heparin 2mg/kg in order to inducing anticoagulation all over the body. Every group separated SMA(superior mesenteric artery) from abdominal cavity and continuously accepted 0.9% Sodium Chloride 10ml(kg·h) using microinjection pump. Intestinal ischemia reperfusion was produced by occlusion of superior mesenteric artery(SMA) for 1h with a clip followed by 2h reperfusion after removal of the clip. Group S separate SMA only. Group I clamped SMA for 1h and then reperfusion for 2h. Group P and E clamped SMA for 1h and received propofol and etomidate at 10mg/(kg·h) and 0.2mg/(kg·h) seperately 5 min before reperfusion. Two hours later, both eyeballs were taken out of rats which were sacrificed by decapitation at the end. The right one and the intestine were kept in 10% formaldehyde, then dyed by hematoxylin eosine(HE) and were observed the morphologic change. The left one was conserved by liquid nitrogen to make homogenate for measuring the activity of SOD and MDA. Data was carried on statistical analysis processing using the SPSS 17.0.Results:1.Pathological change of intestine:compared with group S, intestinal subepithelial space dilat, with epithelium mucosae segregates from lamina propria, in which neutrophilic granulocyte increased and part of villi top breakage in group I. The pathological change of Group P and Group E were slighter than Group I. 2. Pathological change of retina:retina structure is each level clearly demarcated in Group S, the outer nuclear layer arrangement is close, the inside and outside arrange in neat rows rule, and the dividing line is clear. Group I showed that entoretina edema, and ganglion cells scattered with irregular because of the oppression of tissue fluid and outer nuclear layer decomposition and destruction. While Group P and E significantly lessen these changes and reduce retinal edema, and peripheral small vessels open, cells with more rules.3. Compared to group S, the SOD activity is significantly decreased (57.50±10.88) and MDA content increased in Group I (11.60±2.47) (p<0.05). While the SOD activity in group P and E is significantly raised than that in group I (94.50±29.13, 96.25±29.61) (p<0.05), and they can also inhibite the increasing MDA content (5.81±2.92,7.08±1.39) (p<0.05).There was no significantly differences between Group P and E (p>0.05)Conclusion:1.We successfully set up the intestinal ischemia reperfusion rat model and observe that intestinal I/R could lead to injuries of retina. It induced the activity of antioxidase attenuation, the production of OFR increase, the degree of lipid peroxidation enhancement and leaded to changes of above-mentioned correlated index.2.10mg/(kg-h) Propofol and 0.2mg/(kg-h) Etomidate could protect injuries of retina in rats inducing by intestinal I/R. It was possibly dued to improve the activities of antioxidase, clean the OFR, reduce the degree of lipid peroxidation, brain protective effect and so on. There was no significantly differences between Group P and E.
