Objective:To apply metabolomics techniques,The changes of endogenous biomarkers in spleen,kidney and blood of ICR mice aged 3 months and 16 months and the effects of Jinguishenqi pill on markers in 16-month-old mice were compared,To elucidate the biological basis and main metabolic pathway of Jinguishenqi Pill in regulating the aging changes.Method:10 male and 10 female mice of 3 months old were selected as control group and 20 male and 20 female mice of 16 4 months old respectively,Randomly divided into model group and jinkui shenqi pill group.Jinkui group 3was given Jinkui Shenqi Pill,the control group and the model group was given same volume of normal saline as jinkui group.Mice were fasted after 30 days of gastric perfusion.The mice were killed the next day.Blood was taken from their eyeballs,and the spleen and kidney tissues were picked up and was frozen by the liquid nitrogen at a rapid rate.The supernatant fluid was collected from the blood and stored in a refrigerator for reserve at-80 ?.Metabolic markers were detected and identified by UPLC-HDMS,and then retrieved by HMDB and KEGG database and further screened by Progenesis QI 2.1 software.Results:1.Comparison of 3-month control group and 16-month mouse spleen tissue samples,we found that 18 endogenous metabolic differences related to aging in female mice.Among them,13 metabolic differences were down-regulated,5 were up-regulated,and the Jinkui Shenqi Pill regulated 5 of them,and mainly participated in 3 metabolic pathways.Besides,18 endogenous metabolic differences related to aging in male mice.Among them,9 metabolic differences were all up-regulated,9 metabolic differences were down-regulated,and 13 of them were regulated by Jinkui Shenqi Pill,which were mainly involved in 6 metabolic pathways.2.Comparison of 3-month control group and 16-month mouse renal tissue samples,we found that 12 endogenous metabolic differences related to aging in female mice,Among them,8 metabolic differences were down-regulated and 4 metabolic differences were up-regulated.Jinkui Shenqi Pill regulated 6 of them,and mainly participated in 3 metabolic pathways.Also,18 endogenous metabolic differences related to aging in male mice,Among them,15 metabolic differences were down-regulated and 3 were up-regulated.Jinkui Shenqi Pill regulated 14 of them.They mainly participated in 5 metabolic pathways.3.Comparison of blood samples from 3-month-old control group and 16-month-old mice.Nine endogenous metabolic differences related to aging were found in female mice,Six metabolic differences were up-regulated,three metabolites were down-regulated,and Jinkui Shenqi Pill regulated five markers,and they mainly participated in three metabolic pathways.More about that,there were 17 endogenous metabolic differences related to aging in male mice,5 of which were down-regulated in different degrees,Twelve of them were up-regulated,and 11 biomarkers were mainly involved in 4 metabolic pathways.Jinkui Shenqi Pill had the regulative effect on 11 of the 11 biomarkers.Conclusions:1.Compared with the 3-month-old mice,there were age-related changes in the spleen,kidneys and blood of 16-month-old mice.2.Jinkui Shenqi Pill has certain effect on the aging of spleen tissue,kidney tissue and blood of 16 month old mice.3.The effect of Jinkui Shenqi Pill on the 28%markers in the spleen tissue and 72%in the spleen tissue of the male rat was observed in the effect of the marker changes of the age of the mouse.50%markers in female mouse kidney tissue and 83%markers in male mouse kidney tissue were intervened,56%markers in female blood and 65%markers in male mouse blood were intervened.Furthermore,there were more metabolic pathways involved in the aging metabolites in male mice,suggesting that the effect of Jinguishenqi Pill on the aging of male mice was more extensive.
