| Objective: The effect of Danxi gout decoction adder on the activity of xanthine oxidase(XOD)and the expression of URAT1 mRNA in renal tissue of rats with uric acid nephropathy was observed,and the mechanism of reducing the blood uric acid(SUA)and protecting the kidney was discussed.Methods: 60 SPF grade SD male rats were randomly divided into 6 groups: the blank group,the model group,the high dose group,the middle dose group,the low dose group and the group of benzene bromide Malone,10 rats in each group.Except for the blank group,the other groups chose adenine 100mg/kg plus ethambutol hydrochloride 250mg/kg to prepare the uric acid nephropathy rat model by gavage.The control group was given benzbromo Malone,the treatment group was given Danxi gout decoction adder plus high dose,middle dose and low dose of gavage.Continuous administration of 14 d was used to observe the general condition of rats.The changes of blood uric acid(SUA),urea nitrogen(BUN),serum creatinine(SCr),alanine transaminase(ALT)and glutamic pyruvic transaminase(AST)were measured at 2 hours after the last 1 times of administration.After the rats were killed,the liver tissue was taken and the activity of xanthine oxidase(XOD)in liver tissue was detected by ELISA,and the expression of URAT1 mRNA in the kidney tissue was detected by RT-PCR.Results: Animal experiments showed that after using the method of adenine plus ethambutol hydrochloride,the blood uric acid in the model group increased.The blood biochemical test showed that the levels of uric acid(SUA),uric acid nitrogen(BUN)and serum creatinine(SCr)in the rats of 3 doses of Dan Xi Tongfeng Decoction were decreased,and the liver alanine aminotransferase(ALT)and glutamic pyruvic transaminase(AST)were not obvious abnormality.The results of liver detection showed that compared with the model group,3 agents of Danxi gout decoction adder prescription were compared.The activity of xanthine oxidase(XOD)in the liver tissue of rats with uric acid nephropathy was decreased,and the lowest dose group was the most obvious.The renal detection showed that the expression of URAT1 in renal tissue of uric acid nephrotic rats in 3 doses of Danxi gout decoction adder decreased,the most obvious in the low dose group.The levels of serum uric acid and serum creatinine(SCR)and uric acid nitrogen(BUN)increased in benzbromo Malone group.Conclusion: Danxi gout decoction adder can inhibit the activity of xanthine oxidase(XOD)in liver tissue and reduce the production of blood uric acid.The inhibition of renal reabsorption of uric acid(UA)may be achieved by downregulating the expression of URAT1 in the renal tissue,thus reducing the level of uric acid in uric acid nephropathy rats. |
